摘要
研究了一种复合诱变获得雷帕霉素生产菌——吸水链霉菌(Streptomyces hygroscopicus)高产突变株的方法。采用低剂量紫外诱变复合新型常压室温等离子体(ARTP)诱变技术构建雷帕霉素生产菌的诱变育种体系,获得高产雷帕霉素生产菌突变株,经两次复筛,效价较出发菌株提高18.4%。该方法大幅度减少了筛选量,正突变率高达70.59%,可以较短时间内提高雷帕霉素发酵单位,降低生产成本。
We studied a compound mutagenesis method to obtain a high rapamycin-producing mutant strain (Streptomyces hygroscopicus). Moreover, we constructed a mutation breeding system of rapamycin-producing strain using low dose UV mutagenesis and novel atmospheric and room temperature plasma(ARTP),and ob- tained high rapamycin-producing mutant strain,of which the titer increased by 18.4% than original strain by secondary screening. This method significantly reduces the screening quantity, the positive mutation rate can reach up to 70.59% ,improves the rapamycin fermentation unit in a short time,and reduces the production cost.
