壮通饮对脑梗死大鼠内源性神经干细胞的影响 被引量:7

摘要 目的观察壮通饮对脑梗死大鼠内源性神经干细胞(NSC)增殖分化表达的影响。方法将240只雄性大鼠随机分为模型组、假手术组、壮通饮治疗组、正常组,采用线栓法制作大脑中动脉闭塞(MCAO)模型,分别给予不同处理,于术后1d、3d、7d、14d、21d、28d6个时间点分批处死大鼠。用免疫组织化学方法检测各组大鼠梗死区域及相对应区域的神经元核心抗原(NeuN)、胶原纤维酸性蛋白(GFAP)标记的阳性细胞情况。结果模型组脑缺血区域相对应区域可见大量的NeuN、GFAP标记阳性细胞,差异均无统计学意义(P>0.05),与假手术组相比,模型组和壮通饮治疗组大鼠GFAP标记阳性细胞数目增多,NeuN标记阳性细胞数目减少,差异均有统计学意义(P<0.05),与模型组相比,壮通饮治疗组大鼠NeuN、GFAP标记阳性细胞数目均增多,第14天时NeuN、GFAP标记阳性细胞数目达到高峰,差异均有统计学意义(P<0.05)。结论壮通饮可以促进脑梗死后内源性神经干细胞的增殖,并诱导分化为神经元、星形胶质细胞、少突胶质细胞等,但特异性不明显。新分化的神经元有正常的兴奋性,可以和其他神经元形成突触联系,表明壮通饮诱导的神经元可以发挥正常的神经功能。 Objective To observe the effect of Zhuangtong drinking(ZTD)on the proliferation and differentiation of endogenous neural stem cells(NSC)in cerebral ischemia.Methods Two hundreds and forty clean and healthy male Sprague-Dawly(SD)rats were randomly divided into model group,sham operation group,ZTD treatment group and normal group.The middle cerebral artery occlusion(MCAO)model was established by thread inserting method.The rats were killed in postoperative 1 d,3 d,7 d,14 d,21 d,28 d.Immunohistochemistry was used to detect the positive expression of neuronal core antigen(NeuN)and collagen fibrillary acidic protein(GFAP)in the infarcted area and the corresponding area in each group.Results A large number of positive cells marked by NeuN and GFAP in the infarction area and corresponding to the area were observed in model group.Compared with sham operation group,the number of GFAP positive cells increased while the number of NeuN positive cells decreased in model group and ZTD treatment group(P 〈0.05).Compared with model group,the number of GFAP positive cells and NeuN positive cells increased(P 〈0.05),which increased to the top in the 14 th day.Conclusion ZTD can promote the proliferation of endogenous NSC after cerebral infarction,and induced to differentiate into neurons,astrocytes,oligodendrocytes,and so on.But the specificity is not obvious.New differentiated neurons keep the normal excitability and connections with other neurons,which suggestes that the neurons induced by ZTD can play a normal nerve function.
