摘要 目的:优选制备龟甲胶过程中龟甲的炮制方法以及煎煮次数,提高龟甲胶中氨基酸含量。方法:以龟甲胶中L-羟脯氨酸、甘氨酸、丙氨酸、L-脯氨酸等氨基酸含量总和为指标,正交设计法考察不同炮制方法及煎煮次数对制备龟甲胶的影响。结果:龟甲通过蒸制去除腐肉后,不烫淬煎煮3次后的氨基酸含量最高。结论:建立龟甲胶中龟甲的炮制处理方法,确定煎煮次数,提高L-羟脯氨酸、甘氨酸、丙氨酸、L-脯氨酸4种氨基酸含量,稳定龟甲胶质量。 Objective: To optimize the processing method and decocting frequency of tortoiseshell in the process of preparing tortoise-shell glue and improve the amino acid content in tortoise-shell glue. Methods: The sum content of L-hydroxyproline,glycine,alanine,L-proline in tortoise-shell glue as index,orthogonal design method was used to investigate the effect of different processing methods and decocting frequency on the preparation of tortoise-shell glue. Results: The highest acid content was tortoise-shell through steam system,not hot quenching and decoction for 3 times. Conclusion: The processing method was established,the frequency of decoctions determined and the content of L-hydroxy proline,glycine,alanine,L-proline was improved,and the quality of tortoise-shell glue was stabilize.
