摘要
本文报道使用新型混合酶,首次成功分离海萝原生质体的实验结果与培养结果。海萝(Gloilpeltisfurcata)酶解2.5~3h,获得大量成活原生质体。并观察到原生质体的自融。原生质体产率为3×10 ̄6/克鲜藻;成活率为65%。原生质浓厚的细胞经培养,可形成多细胞团或愈伤组织状细胞团,这与小石花菜细胞的培养结果,基本类同。新型混合酶价格低廉、制作简便;在使用上,还有促进细胞分裂、细胞团较长时间存活的优点。新型混合酶的研制和应用,对促进海藻生物工程的研究和海藻资源的开发,对推进21世纪海藻高新技术及其产业的发展,将有重大的现实意义和战略性意义。
篢his paper reports the results of both the experiments and the cultivation ofisolated GlofOpeltis protoplast by successfully using a new type of mixed enzyme(i.e.mixed liquid of viable biological enzyme)for the first time. After Gloiopeltis furcata beingenzymised for 2.5~3hrs, large amounts of survival protoplast can be collected and theself-fusion of protoplast can also be observed.The production rate is 3× 10~6 ind/g freshalgae;the survival rate is 65%,The dense cells of protoplast,after being cuItivated,mayform muIticellular masses or callus tissue-like cell masses,The result of this observa-tion is fundamentally the same as that of the experiment in cultivating Gelidizim divari-catum cells. The new type of mixed enzyme is low in price and simple in production,whiie,in use,it can promote cell divition with the advantage of a longer survival time ofthe cellular masses. The development and application of the new type of mixed enzyme,in the studies on algae biological engineering,in the exploitation of algae resources,and,in putting forward the development of algae science in the 21st century,will be of great,realistic and strategic significance.
