摘要
目的优选巴戟天中水晶兰苷的纤维素酶辅助提取工艺。方法以水晶兰苷提取率为指标,通过HPLC含量测定方法,在单因素考察(酶用量、酶解温度、酶解时间、料液比、提取时间)的基础上,采用二次通用组合设计优化纤维素酶辅助提取的最佳工艺。结果纤维素酶辅助提取巴戟天中水晶兰苷的最佳工艺为:pH值5.0,酶用量0.24%、酶解2.1h、液固比22∶1、提取3次、每次76min,提取率达13.61mg·g^(-1)。结论通过二次通用旋转组合设计所筛选的巴戟天中水晶兰苷纤维素酶辅助提取工艺可显著提高水晶兰苷的提取率,为巴戟天中水晶兰苷的高效利用提供试验依据。
Objective To optimize the enzyme-assisted extraction process of crystal glycoside in Morinda officinalis.Methods Based on a study of the single factors(the enzymatic hydrolysis rate,enzymolysis time,the ratio of material to liquid and the extraction time),the extraction rate of crystal glycoside was optimized using the quadratic general rotary unitized design.The crystal glycoside was used as an index and was determined by HPLC.Results The optimal extraction technology of enzyme-assisted extraction of Morinda officinalis was pH 5.0,enzyme dosage 0.24%,enzymatic hydrolysis of 2.1 h,a liquid-solid ratio of 22∶1,extraction 3 times,76 min/time,and an extraction rate of 13.61 mg·g^-1.Conculsion The experimental results show that the extraction rate of crystal glycyrrhiza can be improved using the quadratic general rotary unitized design,which can provide experimental data for efficient use of Morinda officinalis.
