含食品基质的志贺氏菌标准物质特性值检测及能力验证

Feature Value Detection and Proficiency Testing of Shigella sonnei Reference Materials Containing Food Matrix

摘要 对含鸡肉基质的志贺标物按照《CNAS-GL03能力验证样品均匀性和稳定性评价指南》要求进行均匀性和稳定性的检测,并确定标物的最小取样量。按规定的运输条件送至参加能力验证的实验室,在有效时间内按要求检测完毕,并及时汇报检测结果。本结果根据《CNAS-GL02-2006能力验证结果的统计处理和能力评价指南》进行评价。结果:志贺标物的最小取样量为2 m L(即瓶内全部复水原液),均匀性试验F统计量=1.408<2.424,均匀性良好,可满足能力验证均匀性要求。标物在-20℃和4℃保存10 d的稳定性良好,两组数值的标准偏差分别为0.00741和0.0105,说明-20℃下存放的标物较4℃更稳定,运输时应尽量保持环境温度4℃,在检测时可将其暂时存放于-20℃冰箱。定性能力验证结果满意度为86.7%,参与的30间实验室仅有4间结果不满意。定量能力验证结果满意度为80%。参加的实验室共20间,其中一间检测结果可疑,3间实验室结果不满意。 Detect the stability and uniformity of Shigella sonnei Reference materials in chicken according to the《CNAS-GL03 Guidance on Evaluating the Homogeneity and Stability of Samples Used for Proficiency Testing 》, and to determine the minimum sample quantity. Take the samples to laboratory according the required transport conditions, testing is completed as required in the effective time, and report the results timely. Analysis Results according to the 《Guidance on Statistic Treatment of Proficiency Testing Results and Performance Evaluation》. Results: The minimum sample quantity is 2 m L of(all of the content in the bottle), Uniformity test, the F statistic = 1.408 2.424, so the uniformity is good to meet the ability requirements. Under-20 ℃ and 4 ℃ save 10 d, Reference materials stability is good, the two groups of numerical standard deviation was 0.00741 and 0.00741, respectively, thus-20 ℃ for a standard content more stable than 4 ℃, so keep the environment temperature of 4 ℃ as far as possible when the transportation, before the detection can be stored in-20 ℃. Qualitative ability verification results satisfaction was 86.7%, only four laboratory result is not satisfactory in 30 laboratory. Quantitative ability verification results satisfaction was 80%, one of the test results suspicious and 3 laboratory results not satisfied in the laboratory, a total of 20.
