普罗布考对抗大鼠肾缺血-再灌注损伤影响的研究

Effects of Probucol Against Renal Ischemia-reperfusion Injury in Rats

摘要 目的研究大鼠肾脏缺血-再灌注损伤后,普罗布考修复肾脏病理损伤的影响。方法 30只SD雄性大鼠随机分为假手术组、缺血-再灌注(ischemia reperfusion,IR)组、普罗布考治疗组(普罗布考组),每组10只。假手术组和IR组大鼠每天20ml温开水灌胃,普罗布考组普罗布考250mg/(kg·d)灌胃,1周后制备肾缺血-再灌注大鼠模型。检测3组大鼠的24h尿蛋白定量、尿β-N-乙酰氨基葡萄糖苷酶(β-N-acetylglucosaminidase,NAG)、血清尿素氮(blood urea nitrogen,BUN)、血清肌酐(serum creatinine,Scr)、血清和肾组织中超氧化物歧化酶(superoxide dismutase,SOD),丙二醛(malondialdehyde,MDA)水平;观察大鼠肾组织病理学改变并计算肾脏指数。结果与假手术组比较,IR组、普罗布考组24h尿蛋白定量、尿NAG、BUN、Scr、MDA水平均明显升高,组间比较差异均有统计学意义(P<0.05);血清和肾组织中SOD明显降低,组间比较差异有统计学意义(P<0.05)。与IR组比较,普罗布考组24h尿蛋白定量、尿NAG、BUN、Scr、MDA水平均明显降低,而血清和肾组织中SOD明显升高,肾组织病理损伤明显减轻,肾指数明显改善,组间比较差异均具有统计学意义(P<0.05)。结论普罗布考可以减轻IR大鼠肾功能及肾组织病理损伤,具有保护肾脏的作用。 Objective To investigate the effects of probucol on renal function and renal pathological changes in rats with ischemia-reperfusion (IR) injury. Methods Thirty male SD rats were randomly divided into sham operation group (S group) ,ischemia reperfusion group (IR group) and probucol treatment group (P + IR group), 10 in each group. Rats in S and IR groups were fed with warm water (20 ml/d), and rats in P group were intragastric administration with probu- col [250 mg/(kg.d)]. After i week, rats model of renal ischemia and reperfusion were prepared, and the levels of 24-hour urinary protein, urine β-N-acetylglucosaminidase (NAG), blood urea nitrogen (BUN), serum creatinine (Scr), serum, su peroxide dismutase (SOD) and malondialdehyde (MDA) of all rats were measured. The pathological changes of the kid- neys were observed,and kidney indexs were calculated. Results Compared with the S group,the levels of 24-hour urinary protein,NAG,BUN,Scr and MDA in IR and P + IR groups all increased (P〈0. 05) ,and the levels of SOD in serum and renal tissues were significantly decreased (P〈0. 05). The levels of 24-hour urinary protein,NAG, BUN, Set and MDA in P + IR group were significantly lower than those in IR group, and SOD levels in serum and renal tissues in P + IR group increased, meanwhile, MDA levels were significantly decreased (P〈0. 05). The pathological damage of kidney tissues was reduced, and renal indexs were significantly improved (P〈0. 05). Conclusion Probucol can reduce the renal function and pathological injury of renal tissues in IR injury rats, and play a role in renal protection.
