摘要
目的:探讨通幽汤对食管鳞癌细胞基质金属蛋白酶2、9(MMP-2、MMP-9)及基质金属蛋白酶组织抑制因子(TIMP-1)的影响。方法:体外培养食管鳞癌KYSE150细胞,通幽汤全方作用于细胞,CCK-8实验检测细胞增殖变化,细胞划痕实验观察细胞迁移情况,Transwell法观察细胞侵袭情况,荧光显微镜观察细胞凋亡情况,ELISA法测定食管鳞癌细胞培养上清液中MMP-2、MMP-9、TIMP-1含量,Western-blot法检测细胞MMP-2、MMP-9、N-钙黏联蛋白(N-cadherin)、TIMP-1、E-钙黏联蛋白(E-cadherin)表达量变化。结果:与对照组比较,研究组食管鳞癌细胞迁移速率和侵袭能力下降,MMP-2、MMP-9、N-cadherin蛋白水平降低,TIMP-1、Ecadherin蛋白水平升高。结论:通幽汤可能通过调控食管鳞癌细胞侵袭和转移中的关键分子MMP-2、MMP-9、TIMP-1及钙黏联蛋白的表达,抑制癌细胞的侵袭和转移。
Objective: To investigate the effects Tongyou Decoction on matrix metalloproteinases 2,9( MMP-2,MMP-9) and matrix metalloproteinases tissue inhibitor( TIMP-1) of esophageal squamous cancer cell. Methods: Culturing KYSE150 esophageal squamous carcinoma cells in vitro,joining Tongyou Decoction into the medium of the cells,using wound healing assay to observe the cell migration,using Transwell to observe the cell invasion. ELISA method was used to determinate the MMP-2,MMP-9 and TIMP-1 in the medium of esophageal squamous cancer cells. Results: Tongyou Decoction inhibits the invasion and metastasis of esophageal squamous cancer cells,down-regulating the MMP-2 and MMP-9 levels and up-regulating TIMP-1 levels of esophageal squamous cancer cells. Conclusion: Tongyou Decoction inhibiting tumor cell invasion and metastasis. The mechanism may be the regulation of the key molecules protein of MMP-2,MMP-9 and TIMP-1 of esophageal squamous cancer cell.
