门冬氨酸鸟氨酸颗粒对小鼠急性酒精性肝损伤的影响 被引量:1

摘要 目的研究门冬氨酸鸟氨酸颗粒对小鼠急性酒精性肝损伤的保护作用,并探讨其可能的机制,从而为临床应用提供依据。方法将40只雄性ICR小鼠随机分为正常组、模型组、阳性组(联苯双酯滴丸3.75mg·kg-1)、门冬氨酸鸟氨酸颗粒组(1500mg·kg-1)。除正常组外其余各组每天灌胃52度二锅头酒,建立急性酒精性肝损伤模型;造模同时,灌胃给予受试药,连续17d。末次给药后各组禁食不禁水,12h后摘眼球取血,测定血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)含量。测定肝组织中超氧化物歧化酶(SOD)活性及丙二醛(MDA)的含量,并进行肝脏病理学观察。结果门冬氨酸鸟氨酸颗粒能明显降低酒精性肝损伤小鼠肝组织中MDA含量、血清中ALT、AST水平及肝脏系数,提高肝组织中SOD活性,改善小鼠肝脏病理形态学病变。结论门冬氨酸鸟氨酸颗粒对急性酒精性肝损伤具有一定的保护作用,其机制可能与抗氧化有关。 Objective To study the protective effect of orinithine aspartate granules on acute alcoholic liver injury in mice and to elucidate the possible mechanism.Methods Forty ICR mices were randomly dividedinto four groups:normal group,model group,bifendate dropping pill(3.75 mg·kg-1)group,and orinithine aspartate granule(1500 mg·kg-1)group.Except the normal group,the other groups were administered with 52%liguor.while the acute alcoholic liver injury model was established,each group was respectively administered for 17 days accordingly except the normal control group and model group.After the last administration,the eyeball serum was separated 12 hlater.Alanine transaminase(ALT)and aspartate transaminase(AST)in plasma were detected.The activities of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in liver tissues were determined.Histological changes in the liver were evaluated by hematoxylin-eosin(HE)staining.Results Compared with the model group,orinithine aspartate granules could significantly reduce the content of MDA in liver tissues,ALT levels,AST levels in serum and liver factors in liver tissues,improve SOD activity of liver tissues and obviously alleviate path ological changes of liver tissues.Conclusion Orinithine aspartate granules have protective effect on acute alcoholic liver injury.The mechanism may be related to antioxidation.
