摘要
目的评价银杏叶提取物对重复正加速度暴露高脂血症大鼠主动脉炎症损伤的保护作用。方法雄性SD大鼠50只随机分为5组,对照组、模型组、银杏叶提取物低、中、高(50、100、200mg·kg^(-1))剂量干预组,每组10只。模型组和药物干预组大鼠均给予高脂饲料喂养合并重复正加速度暴露处理,药物干预组大鼠每日灌胃给予各自剂量银杏叶提取物1次。实验持续8周后检测各组大鼠血脂水平;Western blotting及免疫组化法检测大鼠胸主动脉核因子-κB和血管黏附蛋白-1表达以及炎症反应指标。结果银杏叶提取物能减轻重复正加速度暴露合并高脂饮食导致的大鼠胸主动脉炎症反应,抑制核因子-κB p65易位活化,降低血管黏附蛋白-1表达,减少促炎症因子的产生。结论银杏叶提取物能够减轻重复正加速度暴露合并高脂饮食导致的主动脉炎症损伤,其机制可能与其抑制动脉组织核因子-κB p65活化,降低黏附因子水平,减轻炎症反应有关。
Objective To investigate the effect of ginkgo biloba extract(GBE)on inflammation in aortas of high-fat diet-induced hyperlipidemic rats exposed to repeated positive acceleration.Methods Fifty male Sprague-Dawley rats were allocated to five groups by random assignment,including a normal control group and GBE(50、100、200 mg·kg^-1)groups.The rats in control group were fed with common feed,while the rats in other groups were administered with high lipid feed and were exposed to positive acceleration.After eight-weeks of intragastric administration of GBE,the serum was separated,and thoracic aortas were kept.The following indicators were observed:lipid metabolism indicators,inflammation reaction indicators,and expressions of Nuclear factor-κB(NF-κB)and VAP-1.Results GBE significantly decreased the proinflammatory factors,inhibited NF-κB activation and down-regulated VAP-1 expression.Conclusion GBE can suppress the inflammation in aortas of high-fat diet-induced hyperlipidemic rats exposed to repeated positive acceleration by inhibiting NF-κB activation and down-regulating adhesion protein expressions.
