摘要
目的观察糖尿病大鼠脂肪组织对氧磷酶3(Paraoxonase-3,pon3)、磷脂酰肌醇3激酶/蛋白激酶B(PI3K/Akt)的表达及二甲双胍对其干预的作用。方法 60只SD大鼠随机分为正常对照(NC)组、糖尿病(DM)组、二甲双胍治疗(M+DM)组。Western blot检测脂肪组织pon3的蛋白表达,PT-PCR测pon3mRNA的表达。结果 DM组空腹血糖、空腹胰岛素、稳态模型胰岛素抵抗指数、白细胞介素-6(IL-6)、丙二醛(MDA)水平较NC组增高(P<0.05),M+DM组较DM组降低(P<0.05或P<0.01);DM组脂肪组织pon3、PI3K/Akt的表达较NC组降低(P<0.05或P<0.01),M+DM组脂肪组织pon3、PI3K/Akt的表达较DM组增高(P<0.05或P<0.01)。结论脂肪组织pon3表达降低可能参与了糖尿病的发生,二甲双胍上调脂肪组织pon3的表达可能是其改善胰岛素抵抗、糖代谢的部分机制。
Objective To observe the expressions of paraoxonase-3(pon3)and phosphatidylinositol3 kinase/protein kinase B(PI3 K/Akt)in the adipose tissue of diabetic rats and their changes under metformin intervention.Methods Sixty male Sprague-Dawley rats were randomized into the normal control(NC)group,diabetes(DM)group and metformin treatment(M+DM)group.Western blot assay was adopted to detect the expressions of pon3 and PI3 K/Akt.RT-PCR was used to observe the expressions of pon3 and PI3 K/Akt mRNA.Results The levels of FBG,FFA,FIns,IL-6,malondialdehyde and HOMA-IR were significantly higher in DM group,than in NC group(P〈0.05 or P〈0.01).Compared with DM group,those levels in the M+DM group decreased(P〈0.05 or P〈0.01).The expressions of pon3 and PI3 K/Akt in the adipose tissue were lower in DM group than in NC group(P〈0.01),but were higher in M+DM group than in DM group(P〈0.05).Conclusion Lower expressions of Pon3 in the adipose tissue may participate in diabetes.The increase of the expression of pon3 in adipose tissue may be part of the mechanism by which metformin improves insulin resistance and glucose metabolism.
