摘要
目的研究栀子苷对小鼠肾脏缺血再灌注损伤的保护作用及作用机制。方法健康雄性C57小鼠60只随机分为假手术组,肾脏缺血再灌注损伤组,栀子苷低、中、高(5、10、20mg·kg^(-1))剂量组和阳性对照组。用无创血管夹夹闭左侧肾蒂1h,构建肾脏缺血再灌注损伤模型。再灌注24h后,检测血清中肌酐(Cr)、尿素氮(BUN)、核因子-κB(NF-κB)表达水平;评估肾脏过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)和超氧化物歧化酶(SOD)活性,丙二醛(MDA)含量和病理形态;并检测血清和肾脏肿瘤坏死因子(TNF)-α、白细胞介素-1β(IL^(-1)β)和白细胞介素-6(IL-6)的表达。结果与肾脏缺血再灌注损伤组相比,栀子苷组血清中Cr、BUN和NF-κB表达明显降低,肾脏CAT、GPx和SOD活性明显增高,而MDA含量明显减少,病理形态改变明显减轻;此外,肾脏和血清中TNF-α、IL^(-1)β和IL-6的表达明显减少。结论栀子苷对肾脏缺血再灌注损伤有保护作用,其作用机制与减轻炎症反应和抑制氧化应激相关。
Objective To investigate the protective effect of geniposide(GEN)on renal ischemic reperfusion injury(IRI)and its molecular mechanism in mice.Methods Healthy male C57 mice were randomly divided into sham group,renal IRI group,GEN groups at the dosage of(5,10,20)mg·kg^-1 and positive control group.The model of IRI was induced by clamping the left renal pedicle for 1 hwith excision of the right kidney.After 24 hreperfusion,the serum levels of Cr,BUN and NF-κB were determined.The levels of MDA,renal pathology and activities of CAT,GPx and SOD in renal tissues were mesured.Furthermore,the expressions of TNF-α,IL-6 and IL-1βin serum and renal tissue were evaluated.Results Compared with IRI group,the GEN groups had lower levels of Cr,BUN,NF-κB,MDA,TNF-α,IL-6 and IL-1β,but higher activities of CAT,GPx and SOD.Moreover,the morphological changes of renal tissues in the GEN groups were attenuated.Conclusion GEN can protect the kidney against IRI,the mechanism of which is associated with reduced inflammation and suppressed oxidative stress.
