大孔吸附树脂分离纯化翻白草中三萜类化合物及其降血糖作用的研究 被引量:11

摘要 研究大孔吸附树脂分离纯化翻白草中齐敦果酸和总三萜的最优工艺参数。通过比较8种大孔吸附树脂的吸附性能和解吸性能,筛选出AB-8型大孔吸附树脂,正交实验优选大孔吸附树脂分离纯化翻白草中齐敦果酸和总三萜的最佳工艺条件;并通过大鼠体内降血糖实验,评价翻白草三萜类成分的降血糖效果。结果表明,AB-8型大孔吸附树脂分离纯化翻白草中齐敦果酸和总三萜的最佳工艺条件为:上样液浓度为250 g/L,上样量为3 BV,先以4 BV的30%乙醇除杂,再以7 BV的85%乙醇进行洗脱,洗脱流速为2 BV/h;经过纯化后,齐敦果酸和总三萜的纯度分别为2.67%、58.46%,AB-8型大孔吸附树脂对翻白草中三萜类成分有较好的分离纯化效果,优选的方法简单可行;降血糖研究表明,翻白草三萜类成分降血糖作用明显。 To study the isolation and purification technology parameters of oleanlic acid and total triterpenoids from Fotentilla discolor by macroporous resins. Eight macroporous resins were selected with static and dynamic adsorption and desorption assays, and we found that AB-8 macroporous resin was the optimal purification resin. The further orthogonal design was used to study the effects of macroporous resins purification technology with the oleanlic acid and total triterpenoids were served as indexs. And the hypoglycemic effect of triterpenoids was evaluated on diabetes model mouse. In conclusion, the results indicated that the optimal purification conditions were sample mass concentration of 250 g/L, the loaded amount of 3 BV, and the flow rate of 2 BV/h. The collection was obtained by 30% ethanol with 4BV and 85% ethanol of 7BV, respectively. The purity of oleanlic acid and total triterpenoids were 2.67% and 58.46% after the isolation and purification processes. The results indicated that the AB-8 macroporous resin was an ideal resin with the best enrichment for isolating total triterpenoids from Potentilla discolor and the method obtained by orthogonal design was easy to conduct. In addition, the triterpenoids from Potentilla discolor possess anti-diabetic effects of diabetic mice.
