摘要
目的:观察肺腺癌组织中水通道蛋白1(aquaporin1,AQP1)的表达和分布,进一步探讨其临床意义。方法:取2011年1月~2015年6月间病理资料完整,其间未经化疗,行手术切除并经病理确诊的肺腺癌标本40例。同时取癌旁正常组织(远离癌肿组织大于5 cm的正常组织)作为对照40例,应用RT-PCR、SP法、Western blot检测肺腺癌标本中及对照组AQP1的表达和分布。结果:AQP1散在表达于正常肺组织的毛细血管内皮细胞中,而在肺腺癌组织中AQP1表达于肿瘤的血管内皮细胞以及肿瘤细胞膜上、胞浆内。肺腺癌组织中AQP1和mRNA表达水平分为(0.80±0.18)和(0.92±0.23),明显高于正常肺组织,AQP1和mRNA表达水平分别为(0.27±0.11)和(0.30±0.15),P<0.05。结论:AQP1的高水平表达对肺腺癌发生、发展可能有一定的促进作用。
Objective:To observe the expression and distribution of aquaporin 1(AQP1) in lung adenocarcinoma,and to further explore its clinical significance.Methods:40 cases of lung adenocarcinoma specimens were diagnosed by chemotherapy,surgical resection and pathologic examination.The pathology was complete in 2011-01 ~ 2015-06.The expression and distribution of AQP1 in lung adenocarcinoma and control group were detected by RT-PCR,SP method and Western blot.The expression of AQP1 in lung adenocarcinoma was detected by RT-PCR,SP method and Western blot.Results:AQP1 was scattered in the capillary endothelial cells expressed in normal lung tissue,while AQP1 was expressed in the vascular endothelial cells and tumor cell membrane and cytoplasm of lung adenocarcinoma.The expression levels of AQP1 and mRNA in lung adenocarcinoma were significantly higher than those in normal lung tissues(0.80 ± 0.18) and(0.92 ± 0.23),respectively.The expression levels of AQP1 and mRNA were(0.27 ± 0.11) and(0.30 ± 0.15)P 0.05.Conclusion:The high level of expression of AQP1 may play an important role in the development and progression of lung adenocarcinoma
