摘要
目的:建立了高效液相色谱内标法测定微量血清中维生素A含量的方法,继而分析人体维生素A的营养状况。方法:取血清100μl,加入维生素A醋酸酯内标,用1 ml正己烷振荡萃取后离心,取上清液N2吹干,甲醇溶解残渣,HPLC测定。结果:血液中维生素A在0.20~2.00μg/ml之间具有良好的线性关系,维生素A的最低检出限为0.020μg/ml,标准曲线的相关系数r为0.9994,加标回收率为92.2%~107.0%,相对标准偏差(RSD)小于5%;80份样品中维生素A浓度在0.242-0.780μg/ml之间,符合人群平均水平。结论:方法定性准确,灵敏度高,快速简便,适合于微量血清中维生素A的测定,并可以通过测定血清中维生素A含量推算人体的营养状况。
Objective:To establish a method for the determination of vitamin A in trace serum was established by high performance liquid chromatography(HPLC),and then the nutrient status of vitamin A was analyzed.Methods:Take serum 100 μl,add vitamin A acetate internal standard,use 1 ml of n-hexane oscillation to extract and then centrifuge,take the supernatant nitrogen dry,methanol dissolved the residue,HPLC determination.Results:There was a good linear relationship between vitamin A in the blood between 0.20 and 2.00μg/ml,The minimum check for vitamin A is 0.020 μg/ml,The correlation coefficient of the standard curve is 0.9994,The recovery rate was 92.2% ~ 107.0%,and the relative standard deviation(RSD) is less than 5%,The vitamin A concentration in 80 samples was between 0.242 ~ 0.780 μg/ml,which was consistent with the population average.Conclusion:This method is Qualitative accurate,high sensitivity,quick and easy,suitable for the determination of vitamin A in trace serum,and can be measured by the serum vitamin A content of the body's nutritional status.
