正交试验优化黑轮层炭壳菌抗氧化活性成分提取工艺研究

摘要 采用正交试验设计方法,优化黑轮层炭壳菌中抗氧化活性成分的提取工艺。采用超声辅助浸提工艺,选择提取时间、乙醇浓度和固液比为考察因素,以1,1-二苯基-2-苦基苯肼(DPPH)自由基清除率为考察对象,根据单因素试验结果和正交试验设计原理,采用L_(16)(4~3)正交表对黑轮层炭壳菌抗氧化活性成分的超声提取工艺进行研究。在分析各水平因素显著性及其交互作用的基础上,确定最佳提取条件为:提取时间30 min,乙醇浓度100%,固液比为3:100(即浓度30 mg·m L-1)。在此条件下,获得的提取物DPPH抗氧化活性最高,达到60.38%。该提取工艺可为黑轮层炭壳菌中抗氧化活性物质的开发提供一定的理论和应用参考。 To optimize the extraction technology of antioxidant substances fi-om Dcddinia com'entrica by orthogonal experiment. Using ultrasonic assisted soak extraction process, tile effects of ethanol conceutration, solid-liquid ratio, extraction time on scav- enging rate of DFPH radical were inspected. According to the single factor experiment result and the orthogonal experiment de- sign method, L16 (43) orthogonal table was used to study on the ultrasonic extraction technology of antioxidant active compo- nents from the D. concentrica. Based on the analysis on the significance of each level and its interaction, the optimum extraction conditions were as follows: the extraction time was 30 min, ethanol concentration was 100% and the solid-liquid ratio was 1:20 (concentration of 30 mg.mL-1) . kinder these conditions, the DPPH antioxidant activity of the extracts was the highest, reaching 60.38%. The extraction process can provide theoretical and practical reference for the development of antioxidant substances of D. concentrica.
