摘要
目的:探讨利福平对鱼藤酮诱导SHSY5Y细胞损伤的保护作用,并从溶酶体功能的角度阐明其作用。方法:用鱼藤酮诱导SHSY5Y细胞构建帕金森病模型,经利福平预处理,CCK8检测细胞存活率,免疫细胞荧光检测溶酶体的渗透性变化及检测溶酶体pH值的变化。结果:与鱼藤酮组相比,利福平预处理组可明显提高多巴胺神经元的细胞活力;与正常细胞组相比,鱼藤酮可破坏细胞溶酶膜稳定及提高溶酶体内的pH值,利福平预处理可明显改善鱼藤酮介导的溶酶体功能障碍。结论:利福平可能通过改善鱼藤酮诱导的溶酶体功能障碍来保护多巴胺神经元。
Objective : To investigate the effect of rifampicin on lysosomal function during protection against rote- none induced SHSYSY cell. Methods : After pre-treatment with rifampicin, SHSYSY cells were incubated with rotenone for 24h. The viability of SHSYSY cells was measured by CCK8 assay. The lysosomal membrane permeabilization ( LMP ) of cell was measured by detecting the location of Cathepsin D. The lysosome pH was measured under confocal microscopy after dyed with LysoSensorTM Yellow/Blue DND- 160. Results : compared with control group, the viability of SHSYSY cell was obviously decreased, and LMP and lysosome pH increased significantly. However, pre-treatment with rifampicin could reverse such changes. Conclusion: pre-treatment with rifampicin protects rotenone induced SHSY5Y cell via sustaining lysosome structural integrity and acidification.
