摘要 目的观察在Wistar大鼠动脉粥样硬化斑块形成过程中NLRP3及IL-1β的mRNA及蛋白质水平表达变化,了解NLRP3/IL-1β信号通路在动脉粥样硬化形成过程中发挥的作用。方法采取维生素D3联合高脂饲料方法建立动脉粥样硬化模型。并分时间段取材,用实时荧光定量PCR及蛋白免疫印迹法检测NLRP3及IL-1β的mRNA及蛋白水平的表达变化。结果在Wistar大鼠动脉粥样硬化形成过程中,随着时间的延长,NLRP3及IL-1β的RNA和(或)蛋白表达量则逐渐上升。结论该研究提示NLRP3/IL-1β在大鼠动脉粥样硬化发展过程中明显增强,其作用和意义有待研究。 AIM To observe the expressions of mRNA and protein levels in the process of the formation of NLRP3 and IL-1 beta in atherosclerotic plaques of Wistar rats and to explore the role of the NLRP3/IL-1 beta signaling pathway in the formation process of atheroselerosis. METHODS The model of atherosclerosis was established by vitamin D3 combined with a high fat diet. The expressions of mRNA and protein in NLRP3 and IL-1 beta were detected by real-time fluorescent quantitative PCR and Western blotting. RESULTS In Wistar rats, the expressions of RNA and protein of NLRF3 and IL-1 beta increased gradually with the prolongation of time. CONCLUSION The results of this study suggest that NLRP3/IL-1 beta may play an important role in the development of atherosclerosis in rats.
