人脐静脉内皮细胞通过Wnt/β-catenin信号通路响应低切应力刺激 被引量:5

摘要 目的研究不同切应力对人脐静脉内皮细胞(HUVEC)中Wnt/β联蛋白(β-catenin)信号通路的影响及其与动脉粥样硬化的关系。方法采用切应力加载装置对HUVEC分别加载(0、1、15)达因/cm2切应力6、12、18、24 h后,采用实时定量PCR检测该信号通路β-catenin和蓬乱蛋白2(Dvl2)的mRNA水平,免疫荧光技术检测β-catenin、Dvl2的表达和细胞定位。结果低切应力不仅能够促进Dvl2在胞质中的募集,而且能够促进其β-catenin发生核转移。相反,层流切应力能够抑制Dvl2的表达、细胞定位以及β-catenin的核转移。结论 Wnt信号通路参与血管内皮细胞对低切应力的响应。 Objective To investigate the influence of different fluid shear stress( FSS) on Wnt/β signaling pathway in human umbilical vein endothelial cells( HUVECs) and its relationship with atherosclerosis. Methods HUVECs were subjected to different FSS( 0,1,15 dynes/cm2) for various durations( 6,12,18,24 hours) using a shear stress device.Subsequently,real-time quantitative PCR was used to observe the mRNA expression levels of dishevelled 2( Dvl2) andβ-catenin in the Wnt signaling pathway,and immunofluorescence technology to observe their protein expression and localization.Results The low FSS promoted the expression and recruitment of Dvl2 as well as the translocation of β-catenin into the nucleus at the early stage of loading low FSS. Conversely,laminar FSS inhabited their expression and nuclear translocation of β-catenin. Conclusion Wnt signaling pathway participates in the stress response of vascular endothelial cells to low shear stress.
