摘要
目的:评价掺钽羟基磷灰石涂层对骨髓间充质干细胞粘附和增殖的影响,为新型种植体表面涂层的设计及应用奠定实验基础。方法:采用大气等离子喷涂技术在纯钛表面制备掺钽羟基磷灰石涂层复合涂层,利用扫描电镜、X线衍射仪和接触角测量仪表征其表面特性,通过细胞计数、细胞形态及MTT法考察大鼠骨髓间充质干细胞在涂层表面的粘附和增殖情况。结果:掺钽羟基磷灰石复合涂层呈多孔隙结构,具有较高的粗糙度和亲水性。钽的掺杂显著提高了羟基磷灰石与钛基体的结合强度。与钛相比,掺钽羟基磷灰石涂层表面粘附的细胞数目更多,细胞伸展良好,且具有较高的增殖活性。结论:等离子喷涂掺钽羟基磷灰石复合涂层具有良好的细胞相容性,能够促进骨髓间充质干细胞的粘附和增殖。
Objective: To study the effect of tantalum-substituted HA coating on the attachment and proliferation of mesenchymal stem cells, which provide experimental basis for the design and application of dental implant coating materials. Methods: Tantalum-substituted HA coatings were prepared on titanium using atmospheric plasma spraying.The surface topography, composition, roughness and contact angle of tantalum-incorporated HA coating were characterized. Then the adhesion, spread and proliferation of rat bone mesenchymal stem cells were investigated by cells counting, electron microscopic observation and MTT assay. Results: Tantalum-substituted HA coatings have a porous structure,with higher surface roughness and hydrophilicity. The addition of tantalum dramatically increased the bonding strength of HA coating to titanium substrate. There are more cells attaching on tantalum-substituted HA coatings compared to titanium, which spread well and have a higher proliferation activity. Conclusion: Ta-substituted HA coating prepared by atmospheric plasma spraying has good cytocomoatibility, and could promote the attachment and proliferative activity ofmesenchvmal stem cells.
