原钙黏蛋白10基因在胃癌组织的表达及其临床意义

Expression of protocadherin 10 in gastric cancer tissues and clinical significance

目的观察原钙黏蛋白10（PCDH10）在胃癌组织中的表达并探讨其临床意义。方法采用免疫组织化学法检测202例胃癌患者癌组织及其相应的癌旁组织中PCDH10的表达，分析其与患者临床病理特征和预后的关系。结果胃癌组织中PCDH10蛋白的低表达率为63.4%（128/202），显著高于癌旁组织（P＝0.000）。胃癌组织中PCDH10低表达与患者幽门螺杆菌感染状态（P＝0.000）、肿瘤大小（P＝0.000）、淋巴管侵犯（P＝0.001）、浸润深度（P＝0.000）、淋巴结转移（P＝0.000）、远处转移（P＝0.001）及TNM分期（P＝0.000）显著相关，而与患者性别（P＝0.757）、肿瘤诊断时年龄（P＝ 0.312）、肿瘤部位（P＝ 0.512）、肿瘤生长模式（P＝ 0.366）、肿瘤分化程度（P＝ 0.091）和静脉侵犯（P＝ 0.056）无明显相关。Kaplan-Meier生存分析结果显示，PCDH10低表达的胃癌患者与PCDH10正常/高表达患者比较，其总生存期显著缩短，两者比较差异有统计学意义（P＝0.000）。但Cox多因素回归分析结果表明PCDH10低表达不是影响胃癌患者独立预后因素。结论PCDH10在胃癌组织中表达下调，与肿瘤发生进展及患者预后显著相关。

Objective To investigate the expression level of protocadherin 10 （PCDH10） protein in gastric cancer tissues and its clinicopathologic significance.Methods The expression level of PCDH10 protein in 202 gastric cancer tissues and corresponding normal tissues was detected by immunohistochemistry. The association of PCDH10 expression level with clinicopathological characteristics and prognosis of gastric cancer patients was analyzed.Results The expression of PCDH10 protein in gastric cancer tissues was significantly down-regulated in 128 of 202 （63.4%） gastric cancer tissues as compared with their paired normal tissues （P=0.000）. The low expression of PCDH10 protein in gastric cancer tissues was significantly correlated with H. pylori infection （P=0.000）, tumor size （P=0.000）, lymphatic invasion （P=0.001）, invasive depth （P=0.000）, nodal metastasis （P=0.000）, distant metastasis （P=0.001） and TNM stage （P=0.000）, but not with gender （P=0.757）, age at tumor diagnosis （P= 0.312）, tumor location （P= 0.512）, tumor growth pattern （P= 0.366）, tumor differentiation （P= 0.091） and venous invasion （P= 0.056）. Furthermore, Kaplan-Meier analysis revealed that there was significant difference in overall survival （OS） between the patients with low or normal/high expression of PCDH10 in gastric cancer tissues （P=0.000）. However, the results of multivariate Cox analysis showed the low expression of PCDH10 was not an independent factors for gastric cancer patients.Conclusion The expression of PCDH10 in gastric cancer was down-regulated and correlated with tumorigenesis, progressssion and prognosis for gastric cancer.
