长链非编码RNA小核仁RNA宿主基因3在胃癌组织的表达及临床意义 被引量:11

The expression level and clinical significance of long non-coding RNA SNHG3 in gastric cancer

摘要 目的探讨长链非编码RNA(lncRNA)小核仁RNA宿主基因3(SNHG3)在胃癌组织的表达及其临床意义。方法利用癌症基因图集(TCGA)数据分析SNHG3在35例癌旁组织、56例Ⅰ期胃癌组织、123例Ⅱ期胃癌组织、169例Ⅲ期胃癌组织和41例Ⅳ期胃癌组织中的表达水平。利用Kaplan-Meier生存分析探讨SNHG3表达水平与胃癌患者的生存时间之间的关系。应用生物信息学手段分析SNHG3共表达基因影响的生物进程和信号通路。结果lncRNA SNHG3在Ⅰ期胃癌(124.40±10.51,P=0.034)、Ⅱ期胃癌(128.10±9.88,P=0.003)、Ⅲ期胃癌(136.40±7.55,P=0.000)和Ⅳ期胃癌(125.10±13.34,P=0.004)中的表达水平均显著高于癌旁组织(67.16±8.19),并且SNHG3表达量高的胃癌患者的生存时间较短。生物信息学预测显示SNHG3广泛参与RNA剪接、电子呼吸链的传递、mRNA加工、细胞外基质组织、细胞黏附、骨架系统开发、胶原蛋白分解代谢过程、脂肪酸代谢、剪接复合物、癌症通路、磷酸肌醇3激酶(PI3K)-蛋白激酶B(Akt)信号通路、RAP相关蛋白RAP-1A(Rap1)信号通路、Hippo信号通路等在内的多个生物学进程。结论lncRNA SNHG3在胃癌组织中显著高表达,SNHG3表达量高的胃癌患者生存期短。 Objective To explore the expression and the clinical significance of long non-coding RNA (lncRNA) SNHG3 in gastric cancer.Methods TCGA data were used to analyze the expression of SNHG3 in 35 samples of para-cancerous tissues, 56 samples of stage Ⅰ gastric cancer, 123 samples of stage Ⅱ gastric cancer, 169 samples of stage Ⅲ gastric cancer and 41 samples of stage Ⅳ gastric cancer. Kaplan-Meier plotter data was used to explore the association between SNHG3 expression and survival rates in patients with gastric cancer. Bioinformatics was used to analyze the biological processes and signaling pathways which were influenced by SNHG3 co-expression genes.Results SNHG3 was significantly overexpressed in stage Ⅰ gastric cancer (124.40±10.51, P=0.034), stage Ⅱ gastric cancer (128.10±9.88, P=0.003), stage Ⅲ gastric cancer (136.40±7.55, P=0.000), and stage Ⅳ gastric Cancer (125.10±13.34, P=0.004) compared to para-cancerous tissues (67.16±8.19), and gastric cancer patients with high SNHG3 expression had a shorter survival time. Bioinformatics predictions showed that SNHG3 was widely involved in a number of biological processes including RNA splicing, electronic respiratory chain transmission, mRNA processing, extracellular matrix organization, cell adhesion, skeletal system development, collagen catabolism, fatty acid metabolism, splicing complexes, cancer pathways, phosphatidylinositol 3 kinase (PI3K)-protein kinase B (Akt) signaling pathway, Rap1 signaling pathway, and Hippo signaling pathway.Conclusion lncRNA SNHG3 is upregulated in gastric cancer. The survival time of patients with high SNHG3 expression is short.
