Expression of PALB2 in castration-resistant prostate cancer and its clinical significance

摘要 目的:探讨抑癌基因乳腺癌易感基因2(breast cancer susceptibility gene 2,BRCA 2)定位协作(partner and localizer of BRCA2,PALB 2)基因在去势抵抗性前列腺癌(castration-resistant prostate cancer,CRPC)组织中的表达,及其与患者临床病理特征的关系。方法:选取2015年1月—2017年6月天津医科大学第二医院收集的46例CRPC组织标本,其中前列腺腺癌36例,神经内分泌型前列腺癌(neuroendocrine prostate cancer,NEPC)10例,另取同期手术切除的20例前列腺增生(benign prostatic hyperplasia,BPH)组织作为对照。采用免疫组织化学法检测PALB2蛋白的表达情况,并分析其表达与CRPC患者临床病理特征的关系。结果:PALB2蛋白在CRPC患者中的阳性表达率为80.4%(37/46),明显低于BPH患者的95.0%(19/20)(P=0.024);年龄、原发肿瘤局部情况、是否存在区域淋巴结转移以及前列腺特异性标志物(prostate specic antigen,PSA)水平高低对PALB2的表达无明显影响(P值均>0.05),Gleason评分和病理类型影响PALB2的表达阳性率(P值均<0.05)。进一步的相关分析显示,PALB2的表达水平与前列腺腺癌患者Gleason评分呈负相关(γ=-0.376),与病理类型也有相关性(γ=-0.418)。结论:PALB2在CRPC组织中表达阳性率降低,一定程度上可以反映CRPC的增殖与分化程度,参与肿瘤的发生和发展。 Objective: To study the expression level of partner and Iocalizer of breast cancer susceptibility gene 2 (PA1-B2) in castration-resistant prostate cancer (CRPC) tissues, and to explore the relationship of PALB2 expression with the clinical and pathological features of CRPC patients. Methods: Forty-six CRPC specimens including 36 samples from the patients with prostate adenocarcinoma and 10 samples from the patients with neuroendocrine prostate cancer (NEPC), as well as 20 benign prostatic hyperplasia (BPH) specimens (as the control) were collected from January 2015 to June 2017 in Second Hospital of Tianjin Medical University. The expression of PALB2 protein was detected by immunohistochemistry. The relationships of PALB2 expression with the clinical and pathological features of CRPC patients were analyzed. Results: The positive expression rate of PALB2 protein in CRPC tissues was 80.4% (37/46), which was significantly lower than that in BPH tissues [95.0% (19/20), P = 0.024]. The positive expression rate of PALB2 protein was not significantly different in terms of age, primary tumor status, lymph node metastasis and prostate-specific antigen (PSA) level (all P 〉 0.05), but it was significantly different in terms of Gleason score and pathological type of CRPC (both P 〈 0.05). Further correlation analysis showed that PALB2 expression rate was negatively correlated to Gleason score (γ = -0.376) and pathological type (γ = -0.418). Conclusion: The positive reflect the proliferation occurrence and progress rate of PALB2 expression is decreased in CRPC tissues, which can and differentiation of CRTC cells and may be correlated to the of tumors
