摘要
目的研究茶藨子木层孔菌多糖(PRG)在阿尔兹海默症病理损伤中的保护作用。方法利用Aβ_(25-35)诱导PC12细胞损伤作为细胞模型,观察PRG的预保护、修复损伤及抑制凋亡作用,MTT法检测细胞存活率,Annexin-V-FITC双染法检测细胞凋亡率。结果不同浓度PRG对Aβ_(25-35)诱导的神经细胞损伤具有预保护作用,提高存活率,分别为74%(10μg/ml)、81%(50μg/ml)、88%(150μg/ml);不同浓度PRG均能修复细胞损伤,提高存活率,分别为53%(10μg/ml)、55%(50μg/ml)、59%(150μg/ml);不同浓度PRG均能降低Aβ_(25-35)诱导的PC12细胞凋亡,降低凋亡率,分别为28.6%(10μg/ml)、19.3%(50μg/ml)、14.1%(150μg/ml)。结论茶藨子木层孔菌多糖PRG对Aβ_(25-35)诱导的PC12细胞损伤具有明显的保护作用。
Objective To study the protective effect of Phellinus ribis Polysaccharide on the pathologic injury of Alzheimer's disease.Methods The Aβ(25-35) was added to PC12 cells to set up damage modal,the protective,repair and inbibitory effects of PRG were observed,MTT method was used to determine the survival rate and the apoptotic rate was assessed by flow cytometry.Results PRG of different levels all showed pre-protective effect,survival rate was increased,showed 74 percent(10μg/mL),81 percent(50 μg/mL),88 percent(150 μg/mL); PRG of different levels all showed repair effect,survival rate was increased,showed 53 percent(10μg/mL),55 percent(50 μg/mL),59 percent(150 μg/mL); PRG of different levels all can markedly inhibit cell apoptosis,showed 28.6 percent(10 μg/mL),19.3 percent(50 μg/mL),14.1 percent(150 μg/mL).Conclusion PRG has a protective effect in Aβ(25-35) induced damage to PC12 cells.
