摘要
目的观察新葛根芩连汤对链脲佐菌素诱导的2型糖尿病大鼠海马HIF-1α及VEGF表达的影响,探讨新葛根芩连汤改善糖尿病脑内微循环的机制。方法以高脂饮食加链脲佐菌素腹腔注射复制糖尿病模型,分为模型组、新葛根芩连汤大、中、小剂量组,西洛他唑组,给予相应药物干预,另设空白组,8周后处死大鼠,采用PCR法检测海马HIF-1α及VEGF的表达。结果与空白组比较,模型组大鼠海马中HIF-1α明显增加(P<0.05)、VEGF明显增加(P<0.01),与模型组比较新葛根芩连汤可降低HIF1-α(P<0.05)、VEGF(P<0.01)的表达。结论新葛根芩连汤可通过降低糖尿病大鼠海马HIF-1α、VEGF的表达,改善糖尿病大鼠海马缺氧缺血状态,改善脑功能。
Objective To observe the effect of Modified Gegenqinlian Decoction on the expression of HIF-1 and VEGF in hippocampus of type 2 diabetic rats induced by streptozotocin(STZ),and to approach the mechanism of the improvement of cerebral microcirculation in diabetic rats.Methods After 1 month feeding with high fat diet,the diabetes model was established by intraperitoneal injection of STZ and divided into blank group,model group,Modified Gegenqinlian Decoction large,medium and small dose group,and cilostazol group,giving the corresponding drug intervention,the rats were killed after 8 weeks,PCR was used to detect the hippocampal HIF-1 alpha and VEGF expression.Results Compared with the control group,HIF-1 alpha hippocampus in model group were significantly increased(P〈0.05),VEGF increased significantly(P〈0.01),compared with the model group of Modified Gegenqinlian Decoction can decrease HIF1-alpha(P〈0.05),VEGF(P〈0.01) expression.Conclusion The Modified Gegenqinlian Decoction can reduce the expression of HIF-1 alpha and VEGF in diabetic rats hippocampus,improve the hypoxia and ischemia status of hippocampus in diabetic rats and the brain function.
