摘要
目的建立多元素含量的分析方法,探究雄黄内As对大脑组织内源性元素水平的影响。方法建立电感耦合等离子体质谱(ICP-MS)法测定小鼠大脑组织中Na、Mg、K、Ca、Fe、Cu、Zn、Cr、Mn、Co、Ni、Se和As元素含量分析方法,并将所建方法应用到雄黄染毒大脑组织中上述元素的含量测定。该方法使用标准曲线法计算含量,内标法改善基体效应。结果相关系数(r)大于0.9915(K为0.9365);检出限范围为0.0025~22.8344μg/L;仪器精密度RSD小于1.96%;方法重复性RSD小于3.51%;添加不同浓度的混合标准溶液,加标回收率为81.66%~117.05%(K为62.90%),RSD小于5.17%。结论该方法检出限低,干扰少,灵敏度高;雄黄内As可通过血脑屏障在脑组织中蓄积,引起大脑组织内Na、K、Ca、Zn、Cr、Mn、Co、Ni和Se水平的变化,对Mg、Fe和Cu水平的影响不明显。
Objective To establish a method of multi-element analysis to investigate the effect of As in realgar on the level of endogenous elements in brain.Methods An analysis method has been established to test 13 endogenous elements(Na,K,Mg,Ca,Fe,Cu,Zn,Se,Mn,Cr,Co,Ni,As) in the brain of mice with Inductively coupled plasma mass spectrometry(ICP-MS),and then the content of these elements in brain of mice exposed to realgar were determined.The contents of elements were calculated according to their calibration curves,and internal standard method was adopted to reduce matrix effect.Results The experimental results showed that the correlation coefficient(r) was greater than 0.9915(K was 0.9365); the limits detection were 0.0025-22.8344 μg/L; the precision of apparatus with RSD was lower than 1.96%; the method repeatability with RSD was lower than 3.51%; the recovery rate were 81.66%-117.05%(K was 62.90%) with RSD lower than 5.17%.Conclusion This method was applicable to determinate the contents of endogenous elements which had a low limits detection,less interference and high sensitivity.As of realgar can accumulate in the brain through the blood-brain barrier,and cause changes in the levels of Na,K,Ca,Zn,Cr,Mn,Co,Ni and Se in the brain,but with no obvious influence on the levels of Mg,Fe and Cu.
