恐伤孕鼠对仔鼠大脑皮质Reelin、VLDLR及ApoER2mRNA表达影响 被引量:2

摘要 目的观察恐伤孕鼠对仔鼠大脑皮质组织形态学影响及对大脑皮质Reelin、VLDLR及ApoER2的mRNA表达影响。方法将受孕后的雌鼠用随机取数法分为正常组、模型组、补肾组,每组各10只。运用恐吓为主的复合应激法刺激模型组和补肾组孕鼠,正常组不予刺激。同时补肾组每日以中药左归丸浓缩剂2ml/100g体重灌胃,正常组及模型组则根据雌鼠体重予以等量生理盐水灌胃,造模时间为自分组第1天直至产下仔鼠。雌鼠分娩后,仔鼠出生当天每窝随机取1~2只仔鼠断头取全脑,固定,染色,切片,在光镜和电镜观察其组织结构和超微病理改变。另每窝随机取1~2只仔鼠断头取全脑匀浆做定量PCR,观察各组仔鼠大脑皮质Reelin、VLDLR及ApoER2mRNA表达变化。结果模型组仔鼠大脑皮质区光镜下可见细胞较松散,组织结构较模糊,存在散在片状水肿,水肿区域神经细胞数目减少;电镜下可见到多数神经细胞出现细胞器结构异常,如线粒体肿胀,高尔基复合体肥大,部分伴有细胞核固缩。补肾组与正常组比较,仔鼠大脑皮质在形态学上无明显改变。补肾组和模型组仔鼠大脑皮质Reelin、VLDLR及ApoER2mRNA表达均低于正常组(P<0.01),模型组仔鼠大脑皮质Reelin、VLDLR及ApoER2mRNA表达低于补肾组(P<0.01)。结论恐伤孕鼠可以引起仔鼠大脑皮质发育异常,而左归丸通过补肾填精法调节Reelin信号通路从而阻断或降低仔鼠大脑皮质发育障碍损伤的发生。 Objective To observe a fear of injury pregnant rats of young rat cerebral cortex tissue morphology and on the brain cortex Reelin,VLDLR and ApoER2 mRNA expression.Methods After conception of female mice with random access method is divided into normal group,model group,the kidney group,each group of 10 each.Using threats of compound stress method,model group and pregnant rat kidney group,normal group shall not be exciting.At the same time daily pill with traditional Chinese medicine(TCM) left kidney group thickener 2 ml/100 g weight to fill the stomach,the normal group and model group are equal amount according to the weight of female physiological saline lavage,building time gave birth to you on the first day until the young mice.Female mice after childbirth,Wang was born on the day of the random take 1 ~ 2 per litter boy rat beheaded in whole brain,fixed,dyeing,slice,in the light microscope and electron microscope ultrastructural changes and its organizational structure.The other random take 1-2 per litter boy take whole brain homogenate rat break to do quantitative PCR,observe each seed Reelin rat brain cortex,VLDLR and ApoER2 mRNA expression changes.Results(1) Model group young rat brain cortex were visible under cell is relatively loose,the organizational structure is fuzzy,there are scattered in flake edema,edema area reduced numbers of nerve cells; Under the electron microscope visible to most of the nerve cells,abnormal structure of organelles such as mitochondria swelling,Golgi complex hypertrophy,some accompanied by the nucleus pycnosis.Kidney group compared with normal group,Wang rat cerebral cortex has no obvious change in morphology.(2) The kidney group and model group rat brain cortex Reelin,VLDLR and ApoER2 mRNA expression were lower than normal group(P〈0.01),model group rat cerebral cortex Reelin,VLDLR and ApoER2 mRNA expression below kidney group(P〈0.01).Conclusion Fear of injury can cause pregnant rats Wang rat cortical dysplasia,and left to pill by kidney fill fine adjust Reelin signaling pathways that block or reduce the happening of the rat brain cortex injury of developmental disorders.
