清毒提取液对脓毒症大鼠肠黏膜屏障功能保护作用的研究

Protective Effect of Clear Toxin Liquid on Intestinal Mucosal Barrier Function in Rats with Sepsis

目的观察清毒提取液对脓毒症大鼠肠黏膜屏障功能的保护作用并探讨其机制。方法将Wistar大鼠90只,按随机数字表法分为假手术组,模型组,清毒提取液低、中、高剂量组,乌司他汀组。采用盲肠结扎穿孔法(CLP)诱导大鼠脓毒症,制作大鼠脓毒症模型,应用清毒提取液进行干预。观察小肠的病理变化,检测血清肠型脂肪酸结合蛋白(IFABP)、D-乳酸及谷丙转氨酶(ALT),尿素氮(BUN),肌酐(Cr)水平。结果清毒提取液高剂量组肠上皮细胞破坏较模型组轻,小肠绒毛排列较模型组整齐。清毒提取液中、高剂量组与模型组比较,均可以明显降低IFABP、D-乳酸水平(P<0.05);ALT、BUN、Cr模型组与假手术组比较均有数倍的增高。清毒提取液低、中、高剂量组与模型组比较,均可以明显降低ALT、BUN、Cr水平(P<0.05)。结论清毒提取液能够减轻脓毒症对肠上皮细胞的破坏,修复肠黏膜,降低肠上皮细胞的通透性,减轻脓毒症对肝肾功能的损害。

Objective： To investigate the protective effect of clear toxin liquid on the function of intestinal mucosal barrier in rats with sepsis. Methods： 90 Wistar rats were randomly divided into sham operation group,model group,clear toxin liquid low,high dose group,ust statin group. By cecal ligation and puncture（CLP） induced septic rats,the rat model of sepsis,application of clear toxin liquid intervention were used to observe the pathological changes of small intestine,serum IFABP,D-and ALT BUN,lactic acid and Cr level. Results： The results of clear toxin liquid high dose group of intestinal epithelial cell damage was lighter than those of the model group.The intestinal villi of the clear toxin liquid high dose group was more tidy than those of the the model group.Compared with the control group,the levels including the IFABP and the D-levels were reduced significantly in the middle and high dose of the clear toxin liquid group（P〈0.05）. Meanwhile,compared with the sham operation group,the ALT,BUN and Cr in the model group were increased（P〈0.05）. Compared with the model group,those indicators in the clear toxin liquid group were reduced（P〈0.05）. Conclusion： Clear toxin liquid can reduce sepsis on intestinal epithelial cell damage,repair of intestinal mucosa,reduce intestinal epithelial cell permeability and reduce the damage of sepsis on liver and kidney function.
