地榆大黄散对大鼠浅Ⅱ度烫伤模型的影响及机制研究 被引量:4

摘要 目的观察地榆大黄散对大鼠浅Ⅱ度烫伤模型的影响并探索其机制。方法建立大鼠浅Ⅱ度烫伤模型,给药组创面连续外用相应药物14 d。于造模后不同时相点,以硫酸纸称重法测定各组烫伤组织创面愈合率并取其创面全层皮肤制成匀浆,ELISA法测定血管内皮生长因子(VEGF)含量;采血测定其血清肿瘤坏死因子-α(TNF-α)水平及黄嘌呤氧化酶(XOD)活力。结果地榆大黄散可提高不同时相点大鼠烫伤组织创面愈合率及VEGF含量;降低烫伤大鼠血清TNF-α水平及XOD活力,以高剂量组最为明显(P<0.01)。结论地榆大黄散可通过促进大鼠浅Ⅱ度烫伤创面愈合,增加创面组织VEGF表达,抑制烫伤大鼠血清TNF-α水平及XOD活力而发挥对浅Ⅱ度烫伤的治疗作用。 Objective: To observe the influenceof Diyu Dahuang Decoction on superficial Ⅱ scald model of rat,and to explore its mechanism. Methods: Superficial Ⅱ scald models of rat were established. In each treatment group,continuous external use of the corresponding drug in the surface of a wound lasted for 14 days. At different time phase,the wound healing rate of scalded tissues was determined by the method of sulphuric acid paper weighing in each group,and the whole layer skin of the wound was made into homogenate. The content of VEGF was detected by ELISA,and blood was collected to detect the level of TNF-α and the activity of XOD. Results:Diyu Dahuang Decoction could increase wound healing rate and VEGF content of scalded rats at different time points,reduce serum TNF-α level and XOD activity in scalded rats,especially in high-dose group(P〈0.01).Conclusion: Diyu Dahuang Decoction can promote the healing of superficial second degree scald in rats,increase the expression of VEGF in wound tissue,inhibit the level of serum TNF-alpha and XOD activity in scalded rats,and play a therapeutic role in superficial second degree scald.
