葛根素对糖尿病大鼠肾脏的保护作用

Effect of Puerarin on renal protection in diabetic rats

目的研究葛根素(Puerarin)对糖尿病大鼠肾脏的保护作用,并探讨可能的机制。方法用链脲佐菌素(STZ)60 mg/kg腹腔单次注射诱导雄性大鼠,建立糖尿病大鼠模型。随机选取模型成功的大鼠50只,分成5组:模型对照组、葛根素低剂量组、中剂量组、高剂量组、尿毒清颗粒组,每组各10只。再另取10只正常饲养同龄大鼠为正常对照组。每组大鼠分别于给药前及给药后4、8、12、16周测血糖水平。16周后测定各组大鼠24 h尿蛋白定量、肌酐(Scr)、尿素氮(BUN)水平,大鼠肾组织行HE染色,镜下观察病理改变,免疫组化法检测每组大鼠肾组织基质金属蛋白酶-9(MMP-9)的表达水平。结果与正常对照组比较,所有模型组大鼠24 h尿蛋白、Scr、BUN明显升高,镜下肾组织损伤重,免疫组化示肾组织MMP-9表达明显下调。与模型对照组比较,所有葛根素治疗组大鼠24 h尿蛋白、Scr、BUN均有不同程度的降低,镜下肾组织损伤均有不同程度的减轻,免疫组化示肾组织MMP-9表达均有不同程度的上调。结论葛根素可能通过增加MMP-9的表达水平,保护肾脏组织的高糖损害,减缓糖尿病肾病的发生及发展。

Objective To study the effect of Puerarin on renal protection in diabetic rats and explore the possiblemechanisms. Methods The male SD rats were induced by STZ（60 mg/kg）in single intraperitoneal dose to establish thediabetic rats model. Fifty male diabetic rats were randomly divided into 5 groups：the model control group,the low dosegroup,the middle dose group,the high dose group and the uremic granule group,10 rats each. And then the 10 rats withnormal feeding were used as the normal control groups. The levels of blood glucose in each group were measured beforeadministration and after administration at 4 weeks,8 weeks,12 weeks and 16 weeks. After 16 weeks,the 24 h urinaryprotein,blood urea nitrogen（BUN）and serum creatinine（Scr）levels were measured. The pathological changes of renaltissues were observed by HE staining under the microscope. The immunohistochemical method was applied to detect theexpression of MMP-9. Results Compared with the normal control group,24 h urinary protein,Scr,and BUN levels in allmodel groups were significantly increased,renal tissue injurywas serious,and the expression of MMP-9 in renal tissue wassignificantly down-regulated. Compared with the model control group,24 h urinary protein,Scr and BUN levels in allpuerarin-treated groups decreased to different extent and renal tissue injury was reduced to varying degrees. MMP-9 expression levels in renal tissue in all puerarin-treated groups had different levels of increase. Conclusion Puerarin couldincrease the expression of MMP-9,protect the kidneys from high glucose damage,and reduce the occurrence anddevelopment of diabetic nephropathy.
