摘要
目的:观察葡萄糖酸锌(ZnG)和亚硒酸钠(Na2SeO3)对人胚肺成纤维细胞羟脯氨酸(Hyp)合成水平的单独以及联合拮抗作用。方法:用RAW264.7细胞和人胚肺成纤维细胞分别于37℃、5%CO2培养箱进行培养,RAW264.7细胞在细胞生长对数期时加入二氧化硅(SiO2),同时加入不同浓度ZnG、Na2SeO3的溶液,24 h后取RAW264.7细胞培养液与人胚肺成纤维细胞共同培养,作用48 h后测人胚肺成纤维细胞培养液中Hyp含量。结果:ZnG、Na2SeO3单独与联合作用均可以显著降低人胚肺成纤维细胞Hyp合成水平(P<0.05),其中以ZnG 2.25 mg/L和Na2SeO32.0μmol/L浓度的单独与联合作用效果最明显。结论:一定剂量的ZnG、Na2SeO3对SiO2致人胚肺成纤维细胞Hyp合成水平具有单独以及联合拮抗作用。
Objective:To investigate the antagonism of Zinc Gluconate and Sodium Selenite with single and conjunctive forms for synthesis of hydroxyproline of human embryonic lung fibroblasts.Methods: RAW264.7 and human embryonic lung fibroblasts were cultured at 37℃ with carbon dioxide,which concentration was 5%.During the period of logarithm,RAW264.7 were poisoned,sodium selenite and magnesium sulfate were added respectively at the same time.After 24 hours,the RAW264.7 culture fluid and human embryonic lung fibroblasts were cultured together. Hydroxyproline in the culture fluid of human embryonic lung fibroblasts were measured after 48 hours.Results: It was found that Zinc Gluconate and Sodium Selenite did have the effects on sharp decreasing the synthesis lever of hydroxyproline of human embryonic lung fibroblasts.More over,the outstanding effects were taken with the concentration of(2.25 mg/L) for Zinc Gluconate and(2.0 μmol/L) for Sodium Selenite.Conclusion: It suggested that there were single and conjunctive antagonism of Zinc Gluconate and Sodium Selenite for the synthesis lever of hydroxyproline of human embryonic lung fibroblasts induced by silicon dioxide.
