大鼠经γ线照射后T_μ淋巴细胞双肽氨基肽酶活力的定量分析

QUANTITATIVE ANALYSIS OF DIPEPTIDYLAMINOP-EPTIDASE Ⅳ ACTIVITY OF T_μ LYMPHOCYTE IN γ-IRRADIATED RATS

摘要 本文应用显微扫描光度技术观察大鼠经4.5和8.5Gy ^(60)Coγ线全身和局部屏蔽照射后半年外周血T_μ淋巴细胞双肽氨基肽酶Ⅳ(DAPⅣ)活力的定量改变.结果表明,经不同剂量和不同部位照射后,外周血T_μ细胞的DAPⅣ酶活力显示不同程度的降低,各剂量组均低于对照组水平;经相同剂量照射后,全身照射组的酶活力低于屏蔽组,而不同剂量照射后,8.5Gy组的酶活力低于4.5Gy组;根据DAPⅣ酶活力的定量改变所获得的各剂量组恢复顺序与照后半年外周血和骨髓T_μ淋巴细胞数的恢复顺序相同,但是与T_μ细胞数量相比,DAPⅣ酶活力的升高较慢,提示经一定剂量γ射线照射后,细胞内对辐射敏感的酶等生化成分的恢复需要较长时间. In the present paper,the changes of peripheral dipeptidylaminopeptidase Ⅳ (DAP Ⅳ) activity of T<sub>μ</sub>,lymphocyte in rats irradiated by 4.5 and 8.5 Gy <sup>60</sup>Co γ-ray totally and locally for 6 months,were analysed quantitatively by scanning microspectrophotometry.The results showed that the DAP Ⅳ activity of peripheral T<sub>μ</sub> cell decreased differently after 4.5 and 8.5 Gy whole-body and local <sup>60</sup>Co γ-irradiation,and the enzyme activity in all irradiated groups was lower than that of control group.After same dose γ-irradiation,the enzyme activity of whole-body irradiated groups was weaker than that of locally irradiated groups;while after different dose γ-irradiation,the enzyme activity of 8.5 Gy groups was lower than that of 4.5 Gy groups.The order of recovery in all irradiated groups as shown by the quantitative changes of DAP Ⅳ activity coincided with that of T<sub>μ</sub> cell counts of peripheral blood and bone marrow 6 months after irradiation.When compared with the counts of T<sub>μ</sub> cells,however,the DAP Ⅳ activity increased more slowly,suggesting that radiosensitive enzyme components within the cell,such as the DAP Ⅳ,needed a longer time to return to normal level after a definite dose γ-irradiation.
