福氏与宋内氏痢疾菌双价菌苗候选株安全和免疫效果的进一步观察

给恒河猴灌服福氏和宋内氏痢疾菌双价菌苗候选株Fs-18后无任何不良反应.用粪便中分离的该株培养物进行豚鼠Sereny试验,未见有毒力回复突变.Fs-18株经口免疫的恒河猴,对宋内氏毒株和福氏2a(F2a)毒株经口感染具有明显的双重保护效果.用Fs-18株经耳静脉免疫的家兔产生高滴度的抗宋内氏菌和F2a菌的二价血清抗体.DNA探针杂交试验表明,Fs-18株与宋内氏菌侵入质粒的4.1kb Hind Ⅲ片段无同源性.

Bivalent hybrid strain Fs-18 of Sh flexneri 2a and Sh sonnei did not provoke any untoward reactions after oral vaccination of monkeys.Sereny test in guinea pigs showed that the cultures of the hybrid strain from monkey feces did not recover their virulence.Monkeys immunized orally with Fs-18 were protected against both Sh flexneri 2a and Sh sonnei challenges.Rabbits immunized intravenously with Fs-18 produced high titer serum antibodies against both Sh flexneri 2a and Sh sonnei.Hybridization test of DNA probe (4.1kb) with colonies of Fs-18 indicated that there was no homology between Sh sonnei 4.1kb DNA fragment of large plasmid associated with invasion and Fs-18 strain.
