益生性植物乳杆菌K25高密度培养工艺优化 被引量:8

The optimization of high cell density culturing of probiotic Lactobacillus plantarum K25

摘要 采用单因素试验对植物乳杆菌K25的培养基配方以及培养条件进行了优化。确定植物乳杆菌K25的优化培养基配方为:蔗糖15 g/L,葡萄糖15 g/L,大豆蛋白胨26.66 g/L,酵母浸粉13.33 g/L,柠檬酸钠5 g/L,无水乙酸钠5 g/L,K_2HPO_4 2 g/L,MgSO_4 0.2 g/L,MnSO_4 0.05 g/L,吐温80 1.0m L/L;植物乳杆菌K25的最优培养条件为培养温度33℃,同时流加20%的氨水使发酵液pH值保持6.0~6.5,在此条件下静置培养16 h后,活菌数可达5.1×10^(10) CFU/m L。研究结果为植物乳杆菌K25直投式发酵剂的制备提供了理论依据。 In order to study the high density culture conditions of Lactobacillus plantarum K25, single factor test was used to optimize the culture medium and culture conditions. The optimized medium was obtained with sucrose 15 g/L, glucose 15 g/L, soy peptone 26.66 g/L, yeast extract 13.33 g/L, sodium citrate 5 g/L, sodium acetate anhydrous 5 g/L, dipotassium phosphate 2 g/L, magnesium sulfate 0.2 g/ L, manganese sulfate 0.05 g/L, Tween 80 1 mL/L. The fermentation conditions were optimized using this enrichment medium, and the results showed that the viable count of Lactobacillus plantarum K25 reached 5.1×1010 CFU/mL under the conditions that the pH of medium was 6.0-6.5 adjusted with 20% NH4OH, and cultivated statically at 33 ℃ for 16 h. The results provide a theoretical basis for the production of Lactobacillus plantarum K25 lactic acid bacteria starter.
