HPLC法同时测定金刺参九正合剂中4种有效成分

摘要 目的建立HPLC法同时测定金刺参九正合剂中维生素C、没食子酸、儿茶素和表儿茶素的方法。方法采用高效液相色谱法(HPLC),色谱柱为Diamonsil C18(250 mm×4.6 mm,5μm)柱,流动相为甲醇(A)-体积分数为0.03%的磷酸溶液(B),梯度洗脱,流速:0.7 m L·min-1,检测波长:220nm,柱温:30℃。结果维生素C、没食子酸、儿茶素和表儿茶素分别在质量浓度142.8~1 428 mg·L^(-1)、3.500~35.00 mg·L^(-1)、21.60~216.0 mg·L^(-1)和0.900 0~9.000 mg·L^(-1)内呈良好的线性关系(r≥0.999 1,n=6),平均回收率分别为99.8%(RSD=0.3%,n=9)、98.0%(RSD=1.0%,n=9)、99.7%(RSD=0.3%,n=9)和98.0%(RSD=1.1%,n=9)。结论该方法为该制剂的质量控制提供了理论支撑。 Objective To establish a HPLC method for simultaneously determining the contents of vitamin C, gallic acid, catechin and epicatechin in Jinci Shenjiu Zhengheji. Methods HPLC was carried out on Diamonsil C18 column(250 mm ×4. 6 mm,5 μm) ,and the mobile phase was methanol(A)-0. 03% phosphoric acid ( B ) at a flow rate of 0. 7 mL· min^ - 1. The detection wavelength was set at 220 nm, and the column temperature was maintained at 30℃. Results Vitamin C, gallic acid, catechin and epicatechin showed good linear relationships within the ranges of 142.8 - 1 428 mg·L^- 1,3. 500 - 35.00 mg.·L^-1,21.60 - 216.0 mg·L^1 and 0. 900 0 -9. 000 mg·L^1 (r≥0. 999 1 ,n =6) ,and the average recoveries were 99.8% (RSD =0. 3% ,n = 9) ,98.0% (RSD = 1.0% ,n =9) ,99.7% (RSD =0. 3% ,n =9) and 98.0% (RSD = 1. °% ,n =9). Conclusions This method is simple, accurate and repeatable, which can be applied for the quality control of Jinci Shenjiu Zhengheji.
