外源嵌合基因gtfB-ctxB在T2、T3、T4代防龋用转基因生菜植株中的遗传稳定性 被引量:2

Exogenous gtfB-ctxB Chimeric Gene with the Genetic Stability of Transgenic Lettuce Plants in T2,T3,T4 Generation of Dental Caries

摘要 为了观察培育的外源嵌合基因gtfB-ctxB转基因生菜(防龃齿转基因生菜)传代生长情况,初步探索gtfB-ctxB基因在生菜中遗传的稳定性。田地种植T2、T3、T4代外源嵌合基因gtfB-ctxB生菜植株36,64,40株。在不同生长时期,随机选取植物不同组织部位为样本,用聚合酶链式反应技术检测gtfB-ctxB。T2、T3、T4代转基因生菜中均含有外源嵌合基因gtfB-ctxB,并且每代生菜中转基因生菜所占比例基本一样,都超过50%,其中T2代24株,T3代43株,T4代27株。并且3个连续世代植株阳性率间比较,差异无统计学意义(P>0.05)。gtfB-ctxB外源基因在生菜下代中能高效表达,并且具有很好的遗传稳定性。 To observe the genetic stability of gtfB-ctxB gene in lettuce by observing the growth of gtfB-ctxB transgenic lettuce(transgenic lettuce),the gtfB-ctxB lettuce plants were planted in the fields of T2,T3 and T4,with 36,64 and 40 plants. In different growth stages,different tissues of different plants were selected as samples,and gtfB-ctxB was detected by polymerase chain reaction technique. T2,T3,and T4 generation of transgenic lettuce that is exogenous gtfB-ctxB chimeric gene,and the proportion of each generation of transgenic lettuce in the lettuce is essentially the same; more than 50%,24 strains of T2 generation,T3 generation of43 strains,and T4 generation of 27 strains. Furthermore,there was no statistically significant difference among three consecutive generations of plant positive rates(P〉0. 05). gtfB-ctxB exogenous gene can be effectively expressed in the lower generation of lettuce. It has good genetic stability.
