微小RNA在中国汉族人血浆中的分布研究 被引量:2

Study on plasma distribution of microRNA in Chinese Han population

摘要 目的观察中国汉族人血浆中miR-192、miR-206及miR-613的分布情况。方法用反转录-聚合酶链式反应(RT-PCR)法测定116例健康受试者的miR-192、miR-206及miR-613的表达,外加cel-miR-39进行校正,分析不同受试者血浆中miR-192、miR-206及miR-613的表达差异。结果 116例受试者血浆中miR-192、miR-206、miR-613的相对表达量(△Ct)分别为14.01±4.19,17.63±4.42,19.77±4.68,miR-192在血浆中的表达量明显高于miR-206、miR-613(P<0.05);将3组数据分别按四分位数分组后,发现miR-192、miR-206、miR-613各四分组组间差异均有统计学意义(均P<0.05)。结论中国人汉族人血浆中miR-192、miR-206和miR-613呈正态分布且这3种miRNAs在血浆中的分布有显著差异。 Objective To establish a method for extracting and detecting circulating microRNAs(miRNAs) in plasma and to detect the distri- bution of miR - 192, miR - 206 and miR - 613 in Chinese Han people. Methods The total RNA was extracted from 116 healthy subjects plasma by Trizol LS and then tested by reverse transcription -quantitative polymerase chain reaction (RT -qPCR) method. The expression level of circulating miR- 192, miR- 206 and miR- 613 was standardized by cel -39. Results The ACt values of circulating miR - 192, miR -206 and miR -613 in 116 healthy subjects plasma were 14. 01 ±4. 19, 17.63±4. 42 and 19.77 ±4. 68, respectively. The expression of plasma circulating miR - 192 was significantly higher than miR - 206 and miR - 613 (P 〈0. 05). After grouped by quartiles, the differences of circulat- ing miR -192, miR -206 and miR -613 among four groups were statistically significant (P 〈 0. 05 ). Conclusion The expression of circulating miR - 192, miR -206 and miR -613 in Chinese Han people was distributed normally and showed significant differences.
