高糖促进肾小球系膜细胞纤维连接蛋白合成的实验研究

Effects of high glucose on fibronectin synthesis of cultured human glomerular mesangial cells

目的 观察高糖状态下肾小球系膜细胞 (GMC)增殖及纤维连接蛋白 (FN)合成情况。 方法 采用体外 GMC培养 ,噻唑蓝 (MTT)掺入法和细胞计数观察 GMC增殖 ,用 RT— PCR及双抗夹心 EL ISA方法检测 FN的基因及蛋白质水平变化。 结果 高糖对 GMC增殖有抑制作用 ,对 FN的合成呈浓度及作用时间依赖性增高。结论 高血糖在糖尿病肾病的发生发展过程中起重要作用。

Objective To observe the high glucose on proliferation and fibronectin (FN) synthesis of cultured human glomerular mesangial cells. Methods The cultured of human glomerular mesangial cells was established and the cellular proliferation was assessed using MTT incorporation and cell counting, production of FN was analysed by RT PCR and ELISA. Results High glucose depressed MTT incorporation while increased FN synthesis in a dose and time dependent manner. Conclusion Hyperglycemia may make an important role in the development of diabetic nephropathy.
