甲壳素清洁生产过程中柠檬酸钙回收工艺研究 被引量:1

摘要 以红虾虾壳为原料,通过碱性蛋白酶脱蛋白、柠檬酸脱钙制备甲壳素,结合超声辅助法,优化副产物柠檬酸钙回收工艺条件,并对甲壳素成品和柠檬酸钙进行主要质量指标检测。在单因素实验的基础上,通过 Box-Behnken 实验设计对柠檬酸浓度、液料比和超声时间进行优化,得出了柠檬酸钙回收的最佳工艺条件为:柠檬酸浓度10%,液料比12∶1 mL/g,超声时间50 min,柠檬酸钙得率为93.49%±0.07%,甲壳素的得率达到18.93%±0.06%,甲壳素成品质量达到食品级标准,柠檬酸钙成品质量符合我国柠檬酸钙食品添加剂要求。本研究为甲壳素清洁生产过程中柠檬酸钙的回收利用提供科学的理论依据。 In this study, chitin was extracted from Solenocera melantho shell, and the protein was removed by alkaline protease and decalcified by citric acid. Combined with ultrasonic-assisted method, the process conditions for the recovery of calcium citrate were optimized.The main quality indicators of chitin and calcium citrate were tested.On the basis of single-factor test, the concentration of citric acid (%) ,liquid-solid ratio, the uhrasonic time were analyzed and optimized by the Box-Behnken design.The optimized parameters of the extraction technology were obtained as follows:The concentration of citric acid 10%, liquid-solid ratio 12:1 mL/g, the ultrasonic time 50 min.Under these conditions, the yield of calcium citrate was 93.49% ± 0.07 % , the yield of chitin was 18.93 % ±0.06%.The quality of chitin product reached the standard of food grade.The quality of calcium citrate was in accordance with the criterions of Chinese regulations for calcium citrate additives.This study provided a scientific theoretical basis for the process of calcium citrate recovery in chitin clean production.
