Research Progress of FHL2 Regulating Human and Animal Reproduction

摘要 FHL2是FHL家族成员之一,可作为辅助因子与80余种蛋白质互作。FHL2在哺乳动物卵巢和睾丸中均有表达,通过与转录因子WT-1互作参与动物早期性腺分化,并可能通过与SMAD、TGFβ和WNT等信号通路互作,调控卵巢抑制素α亚基基因(INHA)和P450scc的表达,影响卵泡发育和排卵。FHL2表达异常与多种癌症发生相关,包括乳腺癌、卵巢癌、宫颈癌和前列腺癌等。进一步开展对FHL2基因的研究,有助于阐明动物生殖调控和相关癌症发病机制,对辅助生殖技术、提高动物繁殖力的新技术和抗癌药物的研发具有积极推动作用。 FHL2 is a member of FHL protein family and serves as a cofactor interacting with over 80 different proteins. FHL2 could be highly expressed in mammalian ovary and testis, and participate in early gonadal differentiation of animals by interacting with transcription factor WT-1 and regulate the expression of ovarian inhibin a subunit and P450scc by interacting with signaling pathways including SMAD, TGFβ and WNT, thus to affect follicular development and ovulation. Moreover, ectopic expression of FHL2 might be associated with various cancers, including breast cancer, ovarian cancer, cervical cancer and prostate cancer. Further study on FHL2 gene could help clarify animal reproductive regulation and related cancer pathogenesis, and might play an active role in promoting assisted reproductive technologies, new technologies for improving animal fertility and RD of anti-cancer drugs.
