摘要
目的:探索中药复方联合运动疗法治疗慢性心力衰竭(chronic heart failure,CHF)的疗效,并分析其作用靶点。方法:使用冠状动脉结扎法制造大鼠CHF模型,成功后按体质量依次排序,利用数字随机法分为中药组、西药组、模型组、中药加运动组、运动组;按组别给药42 d。利用超声进行其心功能分析,采用实时荧光定量PCR法对大鼠心肌基质金属蛋白酶-2(matrix metalloproteinase,MMP-2)及金属蛋白酶组织抑制因子-2(tissue inhibitor of metalloproteinase,TIMP-2)进行检查。结果:实时荧光定量PCR法检测中药加运动组MMP-2为(1.212±0.146),西药组为(1.605±0.264),两组比较,差异有统计学意义(P≤0.001)。西药组TIMP-2为(1.519±0.269),中药组为(1.327±0.246),两组比较,差异有统计学意义(P=0.009);中药加运动组为(1.839±0.324),优于西药组,差异有统计学意义(P≤0.001);中药组Ⅰ型胶原为(1.013±0.226),西药组为(0.872±0.182),两组比较,差异有统计学意义(P=0.031);中药加运动组为(1.136±0.251),优于西药组,差异有统计学意义(P≤0.001)。结论:中药联合运动疗法治疗慢性心力衰竭大鼠效果显著,可改善改善CHF大鼠心肌重构。
Objective: To explore the curative effect of Chinese herbal compound prescription combined with exercise therapy on chronic heart failure(CHF) and to analyze its target of action. Methods: Rat model of CHF was established by ligation of coronary arteries. According to body weight,the rats were randomly divided into TCM group,western medicine group,model group,TCM plus exercise group and exercise group. The heart function was analyzed by ultrasound,and the expression of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase(TIMP-2) was examined with real-time fluorescence quantitative PCR method. Results: The levels of MMP-2 in Chinese medicine group and plus exercise group and western medicine group were(1. 212 ± 0. 146) and(1. 605 ± 0. 264) respectively by real-time fluorescence quantitative PCR,and there was significant difference between the two groups(P≤0. 001). The levels of TIMP-2 of the western medicine group was(1. 519 ± 0. 269) and that of the Chinese medicine group was(1. 327 ± 0. 246); there was significant difference between the two groups(P = 0. 009). The levels of TIMP-2 of Chinese medicine plus exercise group was(1. 839 ± 0. 324),which was significantly better than that of the western medicine group(P≤. 001). The typeⅠcollagen in Chinese medicine group was(1. 013 ± 0. 226) and that in western medicine group was(0. 872 ± 0. 182),and the difference was statistically significant(P = 0. 031). The levels of TIMP-2 of the exercise group was(1. 136 ± 0. 251),which was significantly better than that of the western medicine group(P≤0. 001). Conclusion: The effect of combination of Chinese herbal compound prescription and exercise therapy in treatment of chronic heart failure rats is significant,which can improve the myocardial remodeling in CHF rats.
