摘要
目的探讨原花青素拮抗砷诱导支气管上皮细胞氧化损伤作用,为揭示原花青素拮抗砷毒性作用提供参考。方法以BEAS-2B细胞为研究对象,分别给予不同剂量As_2O_3(0、5、10和20μmol/L)、葡萄紫原花青素(GSPE)(0、25和50 mg/L)在24和48 h时,利用试剂盒检测氧化应激相关指标[活性氧(ROS)、脂质过氧化物(LPO)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽(GSH)、巯基(-SH)、超氧化物气化酶(SOD)、过氧化氢酶(CAT)和总抗氧化能力(T-AOC)]的表达。采用一般线性模型分析原花青素拮抗砷效应的交互作用。结果与砷染毒组相比,在24和48 h均发现As_2O_3+GSPE50 mg/L组的ROS、MDA和LPO的水平降低(P<0.05),T-AOC、GSH、GSH-Px、CAT、-SH、SOD活性和水平增加(P<0.05),As_2O_3+GSPE25 mg/L组的GSH、T-AOC和SOD升高(P<0.05)。结论在一定浓度范围内,GSPE可以有效地拮抗砷所致的氧化损伤,且随着剂量增加,抗氧化损伤效果提高。
Objective To investigate the effect of the proanthocyanidin on the oxidative damage induced by arsenic in bronchial epithelial cells,and to provide reference for proanthocyanidin antagonistic the toxicity of arsenic. Methods The BEAS-2 B cells were respectively treated with different doses of As_2O_3(0,5,10,20 μmol/L),GSPE(0,25,50 mg/L) for 24 h and 48 h,the expression of oxidative stress indicators(ROS,LPO,MDA,GSH-Px,GSH,-SH,SOD,CAT,T-AOC) were detected by kits. A general linear model was used to analyze the interaction effect between arsenic and GSPE. Results Compared with the arsenic exposure group,the levels of ROS,MDA and LPO decreased at 24 h and 48 h in As_2O_3+ GSPE 50 mg/L group(P〈0. 05),GSH,T-AOC,GSH-Px,CAT,-SH,SOD levels and activity significantly increased in As_2O_3+ GSPE 50 mg/L group(P〈0. 05). GSH,T-AOC and SOD significantly increased in the As_2O_3+ GSPE 25 mg/L group(P〈0. 05). Conclusion GSPE can effectively take against the oxidative damage induced by arsenic,and with the increase of the dose,the better effect of antioxidant damage in a certain concentration range.
