急性心肌梗死与稳定型心绞痛病人高密度脂蛋白胆固醇对内皮功能的影响 被引量:11

摘要 目的研究急性心肌梗死(AMI)与稳定型心绞痛(SA)病人体内高密度脂蛋白胆固醇(HDL-C)对内皮功能影响的差异。方法采用梯度离心法提取正常志愿者(C组,n=28)、急性心肌梗死(AMI组,n=25)与稳定型心绞痛病人(SA组,n=25)体内的HDL,然后检测各组HDL对脐静脉内皮细胞(HUVEC)内皮一氧化氮合酶(eNOS)及其磷酸化位点S1177的表达、一氧化氮(NO)及氧自由基(O_2·^-)水平的影响。结果与C组比较,SA组HDL-C对HUVEC内eNOS、NO、及O_2·^-的影响无统计学意义(P>0.05);与SA组比较,AMI组HDL明显抑制HUVEC内eNOS及其磷酸化位点S1177的表达、降低NO水平和增加O_2·^-含量(P<0.05)。结论 AMI病人体内的HDL通过抑制eNOS/NO/O_2·途径损害内皮功能。 Objective To study the effect of high density lipoprotein(HDL-C)on endothelial function in patients with acute myocardial infarction(AMI)or stable angina(SA).Methods Gradient centrifugation was used to centrifuge HDL from normal volunteers(control group,n =28),AMI patients(AMI group,n =25)and SA patients(SA group,n =25).The effects of HDL on endothelial nitric oxide synthase(eNOS)and its phosphorylation sites at S1177 expression,levels of nitric oxide(NO)and oxygen free radical(O2·-)in human umbilical vein endothelial cells(HUVEC)were detected.Results There was no significant difference in eNOS expression,NO and O2·-levels in HUVES between SA group and control group(P 〉0.05).Compared with SA group,the expression of eNOS and its phosphorylation sites at S117 were inhibited,NO level decreased,and the O2·-content increased in AMI group(P 〈0.05).Conclusion HDL-C damage endothelial function by inhibiting eNOS/NO/O2·-pathway in AMI patients.
