摘要
目的探讨电针后血清对脂多糖(LPS)诱导的软骨细胞IL-1β、IL-6表达的影响。方法将40只两月龄雄性SD大鼠按1∶3随机分为空白组和造模组,造模组采用改良Hulth造模法建立膝骨关节炎模型并随机分为对照组、实验1组和实验2组各10只,实验1组、实验2组分别予电针治疗15 min/d、30 min/d,空白组和对照组正常饲养,共干预12周,疗程结束后,腹主动脉采血,制备实验用血清。选取4周龄健康雄性SD大鼠10只,Ⅱ型胶原酶消化法体外分离培养软骨细胞后分为正常组、模型组、电针15 min组和电针30 min组,使用LPS诱导建立体外培养软骨细胞炎症模型,并同时予大鼠空白组血清、对照组血清、实验1组血清和实验2组血清进行干预,观察4组软骨细胞干预后IL-1β、IL-6表达的变化。结果与正常组比较,模型组软骨细胞IL-1β、IL-6的表达明显升高(P<0.01);与模型组比较,电针15 min组和电针30 min组软骨细胞IL-1β、IL-6表达明显降低(P<0.01,P<0.05)。结论电针后血清能有效抑制软骨细胞炎症因子IL-1β、IL-6的表达,从而抑制软骨细胞炎症反应。
Objective: To investigate the effect of Electro-acupuncture serum on the expression of IL-1 beta and IL-6 in chondrocytes induced by lipopolysaccharide(LPS). Methods: 40 two-month old male SD rats were randomly divided into blank group and model group, The model of knee osteoarthritis was established by improved Hulth model and divided into control group, experiment group 1 and Experiment group 2 randomly, experimental group 1, experimental group 2 were treated with acupuncture treatment of 15 min/d and 30 min/d randomly. The blank group and the control group were reared normally for 12 weeks. After the end of the course,the abdominal aorta was collected, and the experimental serum was prepared. Chondrocytes were isolated from the knee cartilage of10 four-week-old Sprague-Dawley rats of clean grade by mechanical Ⅱ collagenase digestion and were then incubated with 10 ng/m L of LPS to induce apoptosis, While intervention with the serum of blank group, experimental group 1 and experimental group 2, to observe the expression changes of IL-1 beta and IL-6 in the chondrocytes. Results: Compared with the blank group, the expression of IL-1 beta and IL-6 in the chondrocytes in the model group was significantly increased(P〈0.01); Compared with the model group, the expression of IL-1 beta and IL-6 in the chondrocytes in the EA 15 min group and the EA 30 min group was significantly decreased(P〈0.01, P〈0.05). Conclusion: Electro-acupuncture serum can effectively inhibit the expression of inflammatory factors IL-1 beta and IL-6 in chondrocytes, thus inhibiting the inflammatory response of chondrocytes.
