红花黄色素对糖尿病肾病的保护作用 被引量:18

摘要 目的观察红花黄色素对糖尿病肾病模型的治疗作用和相关机制。方法将60只SD大鼠随机分为正常组、模型组、胰岛素组、红花黄色素高剂量组和红花黄色素低剂量组。除正常组外均采用小剂量链脲佐菌素(STZ,30 mg/kg)腹腔注射+高脂饮食造模12 w,制备糖尿病肾病模型,每只大鼠的胰岛素剂量为0.5 U/d;腹腔注射红花黄色素(高剂量组:40 mg/kg、低剂量组:20 mg/kg),正常组和模型组以等量的生理盐水注射,持续4 w,给药期间每2 w测定空腹血糖(FBG)和血清中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-18,同时收集动物24 h尿液,测定尿蛋白排泄率、尿蛋白(UPRO)等指标;给药4 w后,采用实时定量聚合酶链反应(real-time PCR)测定肾脏中NLRP3、Caspase1 mRNA表达。结果给药之后,与模型组相比,红花黄色素高剂量组和胰岛素组组均能显著降低FBG、UPRO、TNF-α、IL-1β、IL-18等指标;同时,红花黄色素能够下调Caspase1和NLRP3 mRNA在肾脏中的表达(P<0.05)。结论红花黄色素可能通过下调Caspase1和NLRP3 mRNA的表达减轻糖尿病肾病的炎症损伤。
