白藜芦醇调控NF-κB/RIP2信号缓解大鼠膀胱癌的作用 被引量:3

摘要 目的研究白藜芦醇(Resveratrol,RES)通过调节NF-κB/RIP2信号通路来治疗和缓解大鼠膀胱癌的保护作用。方法大鼠构建膀胱癌模型后,白藜芦醇(20 mg/kg)和生理盐水连续灌胃14 d。HE染色法观察膀胱组织形态;Western blot法和实时PCR检测NF-κB、RIP2、Caspase 3和Bcl-2蛋白含量和mRNA含量的表达。结果与对照组相比,模型组和实验组大鼠NF-κB、RIP2和Caspase 3 m RNA和蛋白含量显著升高,Bcl-2 m RNA和蛋白含量显著降低,差异具有统计学意义(P<0.05);与模型组相比,实验组大鼠NF-κB、RIP2和Caspase 3 m RNA和蛋白含量显著降低,Bcl-2 m RNA和蛋白含量显著升高,差异具有统计学意义(P<0.05)。结论 b白藜芦醇能够有效缓解和抑制膀胱癌细胞的生长,且NF-κB/R1P2信号通路在此过程中发挥重要作用。 Objective To investigate the protective and therapeutic effects of resveratrol (RES) on bladder cancer in rats by regulating NF-KB/RIP2 signaling pathway. Methods Rats were established bladder cancer model, resveratrol (20 mg/kg) and saline for 14 days. The morphology of bladder tissue was observed by HE staining. The protein and mRNA expression of NF-kB, RIP2, Bcl-2 and Caspase 3 were detected by Western blot and Real-Time PCR. Results Com- pared with the control group, the NF-KB, RIP2 and Caspase 3 mRNA and protein expressions in the model group and the experimental group were significantly increased, while the contents of Bcl-2 mRNA and protein were significantly decreased (P〈0.05). Compared with the model group, the NF-KB, RIP2 and Caspase 3 mRNA and protein expressions in the experi- mental group were significantly decreased, while the contents of Bcl-2 mRNA and protein were significantly increased in the experimental group (P〈0.05). Conclusion Resveratrol can effectively relieve and inhibit the growth of bladder cancer cells, and NF-kB / RIP2 signaling pathway plays an important role in this process.
