摘要
目的建立了同时测定生姜超临界CO_2萃取物中6-姜酚、8-姜酚、6-姜烯酚、10-姜酚、8-姜烯酚、10-姜烯酚的方法。方法采用HPLC-DAD法,Kromasil C_(18)柱(250 mm×4.6 mm,5μm),0.1%磷酸-乙腈作为流动相,梯度洗脱,流速1 m L·min^(-1),检测波长282 nm,柱温35℃。结果 10-姜酚、10-姜烯酚、8-姜酚、8-姜烯酚、6-姜酚、6-姜烯酚进样量分别在0.0853~1.06、0.192~2.40、0.115~1.44、0.064~0.815、0.084~1.05、0.115~1.44μg时与色谱峰面积呈线性型关;加样回收率在99.24%~101.32%之间,RSD在0.97%~1.92%之间。结论建立的测定方法专属性较好,为生姜超临界CO_2萃取物的质量控制提供了参考。
Objective To detecting the content of 6-shogaol, 8-gingerol, 6-shogaol, 10-gingerol, 8-gingerol, 10-shogaol in supercritical extraction of ginger. Methods Using HPLC-DAD method, Kromasil Cis column (4.6 mm × 250 mm, 5μm) with mixture of acetonitrile and 0.1% phosphoric acid as mobile phase. Choosing gradient elution method at flow rate of 1.0 mL·min-1 with detection wavelength of 282 nm. The column temperature was 35 ℃. Results 10-gingerol, 10-shogaol, 8-gingerol, 8-shogaol, 6-gingerol, 6-shogaol showed good linear relationships within the ranges of 0.0853-1.06, 0.192-2.40, 0.115-1.44, 0.064-0.815, 0.084-1.05, 0.115-1.44 μg. Average recoveries were within 99.24 %-101.32 % (RSD: 0.97 %-1.92 %). Conclusion The established HPLC assay was accurate and reference was provided for the quality control of the CO2 supercriticel extract of ginger.
