三维荧光二阶校正法同时快速测定人体液中两种非甾体抗炎药萘普生和二氟尼柳 被引量:6

Fast and Simultaneous Determination of Naproxen and Diflunisal in Biological Fluids Using Excitation-emission Matrix Fluorescence Coupled with Second-order Calibration Method

摘要 使用了一种快速、绿色、高效的分析策略,即采用三维荧光结合二阶校正方法同时测定人体血清和尿液中的两种非甾体抗炎药萘普生(NAP)和二氟尼柳(DIF)。即使分析物之间以及分析物和未知干扰物之间的光谱存在严重重叠现象,基于交替归一加权残差(ANWE)算法的二阶校正方法仍能对其进行解析,并获得可靠的定性定量结果。在血清和尿液中,NAP的平均回收率分别为(95.6±2.1)%和(90.2±1.2)%,DIF的平均回收率分别为(100.0±0.8)%和(94.2±1.6)%。另外,使用品质因子,如灵敏度(SEN)、选择性(SEL)、检测限(LOD)和定量限(LOQ),评估了该方法的准确性。所得结果均令人满意,表明所提出的方法能够用于人体液中NAP和DIF的准确、快速定量分析。 In this work, a fast, green and effective analysis strategy that the excitation-emission matrix fluorescence(EEMf) coupled with second-order calibration method based on alternating normalization-weighted error(ANWE)algorithm was developed for simultaneous determination of NAP and DIF in human plasma and urine. Although the fluorescence spectra of target analytes overlapped and unknown interferences coexisted in the matrices, reliable qualitative and quantitative results were obtained by the proposed method. The obtained average recoveries were(95.6±2.1)% and(90.2±1.2)% for NAP,(100.0±0.8)% and(94.2±1.6)% for DIF in serum and urine, respectively. In order to evaluate the performances of the proposed method, the figures of merit such as sensitivity(SEN), selectivity(SEL), limit of detection(LOD) and limit of quantification(LOQ) were investigated. The satisfactory results indicated that it may be promising as an effective strategy for quantifying NAP and DIF in biological fluids rapidly and accurately.
