上调miR-29a对婴幼儿血管瘤内皮细胞生物活性的影响

The effect of miR-29a up-regulation on the biological activity of infantile hemangioma endothelial cells

摘要 目的探讨上调mi R-29a对增殖期婴幼儿血管瘤内皮细胞增殖、凋亡和侵袭能力的影响。方法自2016—2017年采用组织块贴壁法行体外细胞培养,以CD31磁珠分离培养婴幼儿血管瘤来源的内皮细胞;通过CCK-8试剂检测细胞增殖能力的变化;通过Annexin V-FITC/PI流式细胞术检测细胞凋亡的变化;以Transwell侵袭实验检测侵袭能力的变化。结果 mi R-29a过表达慢病毒转染细胞72 h后,慢病毒转染组的细胞凋亡率达到(12.54±0.02)%,对照组细胞的凋亡率为(0.29±0.01)%,其较对照组差异有统计学意义(P<0.05)。而上调了细胞内的mi R-29a表达水平后,对细胞增殖及侵袭能力没有显著性影响。结论上调细胞内mi R-29a表达水平对细胞凋亡具有显著性影响,提示mi R-29a可能通过改变内皮细胞凋亡比例来调节瘤体生物活性,并在血管瘤增殖期向消退期转变中起到一定的作用。 Objective To explore the effect of mi R-29 a up-regulation on proliferation, apoptosis and invasion ability of infantile hemangioma endothelial cells in the multiplication period. Methods From 2016 to 2017 the cells were cultured in vitro by the tissue mass adherence method, and endothelial cells derived from the infantile hemangioma were extracted with CD31 magnetic beads. The level of mi R-29 a in cells was up-regulated through mi R-29 a overexpression of lentiviral transfection. Cell proliferation was examined by the CCK-8 method. Apoptosis of Hem ECs was detected by Annexin V-FITC/PI flow cytometry, and the invasion ability was examined by Transwell assay. Results Seventy-two hours after mi R-29 a overexpression lentiviral transfection, the cell apoptotic ratio was(0.29 ±0.01)% in the lentiviral transfection group, and(12.54 ± 0.02)% in the control group. The difference was statistically significant. After upregulation of mi R-29 a expression in cells, there was no significant influence on the proliferation or invasive ability of the cells. Conclusion Up-regulation of mi R-29 a has a significant effect on apoptosis, which suggests that mi R-29 a can modulate biological activity of tumors by changing the apoptosis proportion of endothelial cells. It plays a certain role in the transition from the multiplication period to the regression period of hemangiomas.
