肥胖大鼠袖状胃切除术后脂肪组织激素敏感性脂肪酶及肉碱脂酰转移酶-1表达变化 被引量:1

Expression of hormone sensitive lipase and carnitine acyl transferase -1 in obese diabetic rats after sleeve gastrectomy

摘要 目的观察袖状胃切除术(sleeve gastrectomy,SC)对肥胖大鼠(Zueker大鼠)脂肪组织激素敏感性脂肪酶hormone sensitive lipase,HSL)及肉碱脂酰转移酶-1(carnitine acyl transferase-1,CAT-1)表达的影响,探讨其减重机制。方法30只10周龄雄性Zucker大鼠按随机数字表法随机分为手术组、假手术组、饮食配对组,每组10只。术后6周处死大鼠,取腹膜后脂肪组织,应用RT.PCR及Westernblot技术检测各组HsL和CAT-1基因mRNA及蛋白表达水平。结果手术组术后6周体重较其他2组明显降低,分别((250.0±5.8)、(370.0±10.0)及(310.0±9.6)g(P〈O.05),HSL和UCP-1基因mRNA及蛋白表达均明显升高(P〈0.05)。结论SG能使肥胖大鼠脂肪组织脂解作用关键基因HSL和CAT-1的表达增强,促进脂肪水解,增加能量消耗,是其减雷的莺喜机制岁一. Objective To investigate the expression of hormone sensitive lipase (HSL) and carnitine a- eyl transferase-1 (CAT-l) in Zueker rats after sleeve gastrectomy and to discuss the weight loss mechanism. Methods 30 male Zucker rats aging 10 weeks were randomly divided into 3 groups: the operation group (10 rats), the sham operation group (10 rats) and the diet-pairing group (10 rats). The rats were decapitated to re- trieve the retroperitoneal adipose, mRNA and protein expression of HSL and CAT-1 gene of were detected by RT- PCR and Western blot. Results As for the operation group, the weight decreased significantly after the operation comparing to the other two groups ((250±5.8) g, (370±10.0) g, (310±9.6) g, P〈O.05). mRNA and protein ex- pressions of HSL and CAT-1 gene were all significantly higher in the operation group (P〈0.05). Conclusion SG can up-regulate the expressions of lipolysis gene HSL and CAT-1 in adipose in Zucker rats, promoting fat hydrol- ysis and increasing the energy expenditure, which is one of the important mechanisms of weight loss.
